CAV1 plays a key role in stem cells physiological activity and burn wound healing according to references and our previous study. However, the molecular mechanism is still unknown. We suppose CAV1 overexpressed in ESCs may control the differentiation of ESCs to promote burn wound healing by targeting Notch signal pathway. In this project, the effects of CAV1 in the expression of Notch signal pathway, the ESCs’ differentiation and burn wound repair will be observed both in vivo and in vitro. A lentivirus-based gene transfection technology will be performed to construct a model of ESCs that overexpresses CAV1 to study ESCs proliferation,differentiation, adhesion,migration, to illustrate the effects of CAV1 in would healing and Notch signal pathway and the mechanism of CAV1 in accelerating burn wound healing. Results may provide a new therapeutic treatment and theoretical basement to promote burn wound healing.
CAV1通过信号通路参与调节干细胞的生理活动。文献和我们前期研究发现表皮干细胞(ESCs)过表达CAV1能促进烧伤创面愈合,但其分子机制仍不完全清楚。我们结合研究基础推测CAV1过表达的ESCs可能通过靶向Notch信号通路调控ESCs的增殖分化促进烧伤创面的修复。本项目拟通过离体和在体模型系统的研究CAV1对Notch信号的时空表达、ESCs的增殖分化及对烧伤创面愈合的影响;基于慢病毒转染技术构建CAV1过表达的ESCs离体研究其细胞增殖、分化、黏附与迁移能力的变化,在体验证对小鼠烧伤创面愈合的影响及Notch信号的调控作用,探讨CAV1、Notch信号通路与ESCs增殖分化和烧伤创面修复的相互关系,阐明CAV1改善ESCs介导的烧伤创面修复作用的相关分子机制,为临床上促进烧伤创面修复,改善创面愈合质量开拓新的思路和提供相关理论依据。
烧伤救治的主要目的是全力挽救生命、迅速封闭创面、恢复皮肤功能。鉴于表皮干细胞在创面愈合中的重要作用,我们课题组一直专注于表皮干细胞(ESCs)对创面愈合的研究,文献和我们前期研究发现ESCs过表达CAV1能促进烧伤创面愈合,但其分子机制仍不完全清楚。我们结合研究基础推测CAV1过表达的ESCs可能通过靶向Notch信号通路调控ESCs的增殖分化促进烧伤创面的修复。本研究在烧伤差异基因集的生物信息学分析的基础和指引下,利用在体皮肤创面和离体ESCs分化模型,分别从CAV1及Notch信号通路成员的时空表达特点、干预小鼠深II°烧伤创面愈合模型在体验证CAV1过表达的ESCs对Notch信号的时空表达、ESCs的增殖、分化转归及对创面修复和瘢痕形成的影响,系统阐明CAV1过表达的ESCs干预的烧伤创面愈合过程中的分子机制,将深化CAV1对创面修复分子机制的认识,为临床加快烧伤创面愈合速度,改善愈合质量开拓新思路和提供理论依据,并可能为治疗烧伤创面提供新的治疗靶点。
