Based on the coke oven cohort study carried out in 1980-1990's in China, 3 molecular epidemiological study design were used in this project including cohort study, nested case-control study and cross-sectional study. The relationship between coke emission exposure and early biological effects (DNA damage and chromosomal damage) and risk of lung cancer was investigated, and the effect modification of genetic polymorphisms of several xenobiotic-metabolic enzymes (including GSTM1, NQO1, NAT2 and mEH) was found. The gene-environment interaction and gene-gene interaction are the mechanic models for the carcinogenesis in lung. The results of this project partially provide the theoretical basis for screening high-risk subgroup and prevention.
本课题旨在研究焦炉工肺癌发生过程中的环境基因交互作用机制。通过对职业性接触多环芳烃类化合物工人的队列研究,从多水平上探讨职业性暴露多环芳烃、基因多态性与工人的早期遗传学效应以及职业性肺癌之间的关系。探讨焦炉工肺癌发生过程中的环境基因交互作用的分子机制。为筛检高危人群和早期受罹者,并采取对性的控制措施提供理论基础。.
