Esophageal squamous cell cancer (ESCC) ranks fourth among cancer related death in our country. Metastasis is the primary cause of the poor prognosis. NM IIA is actin-based molecular motor protein, its function depends on the modulation of its assemble state. Currently there is still debate about the effect of NM IIA on metastasis, and proteins that promote NM IIA assemble and disassemble are all reported with increased expression. We speculate that the reason may depend on the expression amount of NM IIA in the studied tissues and that these proteins might modulate different sites of NM IIA. In our previous research, we found that NM IIA expression was elevated in ESCC tissues and cells and NM IIA can promote ESCC migration. In the current study, we will first study the expression amount of NM IIA and its isoform NM IIB in ESCC, and their effect on the cellular biology of ESCC, investigate the effect of disassemble-promoted protein S100A4 and assemble-promoted phospho-MLC on the assemble status , distribution, and cellular function of ESCC. All these study will provide a new direction for in-depth study of the mechanism of ESCC metastasis.
食管鳞癌是我国死亡率居第四位的恶性肿瘤,转移扩散是其预后不良的首要原因。NM IIA是能够结合并移动肌动蛋白的分子马达蛋白,其功能依赖于对其装配状态的调节。目前关于NM IIA在细胞迁移中的作用尚存争论,且促使NM IIA装配及促使NM IIA去装配的蛋白在肿瘤组织及细胞中都呈高表达,我们推测其可能是因为这些组织中NM IIA表达量不同及这些蛋白质可能作用于细胞内不同位置的NM IIA。前期研究发现NM IIA在食管鳞癌组织及细胞高表达,且促进细胞迁移。因此在本项目中首先检测NM IIA和其异构体NM IIB在食管鳞癌细胞中的含量,研究它们对食管鳞癌细胞生物学行为的影响,观察能促进NM IIA去装配的S100A4及促进其装配的磷酸化轻链对食管鳞癌细胞内NM IIA装配状态、分布位置进而研究其对细胞功能的影响。本研究的实施,将为阐明NM IIA促进食管磷癌转移机制提供新的理论基础。
