Dry eye, as a common disease in ophthalmology, is becoming a severe threaten to people life. The chronic inflammation on ocular surface is a main pathogenesis of dry eye and has been proved in many studies. However, there is still few method to control the inflammation so far. Macrophage and its polarization has been found to exhibit an immune modulation function in many other diseases. Moreover, our early study showed that macrophage was involved in the chronic inflammation of dry eye. So, we assume that the anti-inflammation M2 macrophage may help to control dry eye. In this research, we will study the function of M1 and M2 macrophage in dry eye chronic inflammation and their impact on cornea epithelial cell, explore the mechanism and influence factor of macrophage polarization in dry eye model, and observe the effect of M2 polarization and M2 condition medium on the ocular surface damage in dry eye. Base on the research result, we hope to illuminate the function of macrophage and the possibility to improve dry eye, so as to manage dry eye better.
干眼作为一个眼科常见疾病,正影响越来越多人的生活质量。眼表的慢性炎症反应是干眼的主要发病机制之一,虽然已有较多研究证实,但目前尚无有效的干预方法。巨噬细胞的特有的极化方式已在多种疾病中发现具有免疫调节作用,我们的前期研究结果也表明干眼的眼表慢性炎症反应中有巨噬细胞的参与,因此提出具有抗炎作用的M2型巨噬细胞能够改善干眼的假设。通过研究干眼模型的慢性炎症反应中M1型和M2型巨噬细胞的作用及其对角膜上皮细胞的影响,探讨干眼模型中M1型和M2型巨噬细胞极化的调控机制及影响因素,观察诱导巨噬细胞向抗炎的M2型极化及M2型巨噬细胞条件培养基对改善干眼模型中眼表损伤及炎症反应的作用,阐明巨噬细胞在干眼中的作用及M2型巨噬细胞对改善干眼的效果,为临床上干眼的治疗和管理提供新的靶点。
