Age-related cataract (ARC) is the leading cause of irreversible blindness in the world and its pathogenesis is not clear. Several studies confirmed that oxidative stress-induced lens epithelial cells (LECs) apoptosis is closely related to ARC formation. Our study has found that nuclear factor erythroid-derived factor 2-related factor 2 (Nrf-2)/ heme oxygenase-1 (HO-1)/ carbon monoxide (CO) signaling pathway plays a critical role in preventing LECs from oxidative and endoplasmic reticulum (ER) combined stress. Recently, we confirmed CO strengthened the anti-oxidative and anti-apoptotic ability LECs. In this study, the clinic specimens of ARC patients will be tested with a correlation analysis. The Nrf2/HO-1/CO axis will be induced by cell-penetrating peptides (CPPs) + drugs in both cell and animal models (mutant cataract mice and selenite cataract rabbit). Therefore, the protective mechanism of Nrf2/HO-1/CO axis on oxidative -ER stress and ARC formation will be dissected.
年龄相关性白内障(ARC)是世界首位致盲性眼病,其发病机制尚不清楚。氧化应激介导的晶状体上皮细胞(LECs)凋亡与ARC的发生发展密切相关。本课题组前期研究发现核因子E2相关因子2(Nrf-2)/血红素加氧酶-1(HO-1)/一氧化碳(CO)信号通路参与氧化应激和内质网的联合应激,在保护LECs消除损害方面具有重要作用。本课题组在成功探究CO在抗氧化应激及抗细胞凋亡中的作用基础上,采集ARC患者标本进行Nrf2/HO-1/CO通路检测,明确通路与ARC相关性;构建细胞穿膜肽(CPPs)—药物复合体,干预信号通路,在细胞和不同动物模型(包括化学诱导型和基因修饰型)层面探究Nrf2/HO-1/CO通路在ARC发病过程中的保护机制。为精准研发有效的ARC治疗药物奠定基础。
年龄相关性白内障是世界性第一位致盲性眼病,其发病机制尚未完全阐明。目前证实氧化应激与其发生密切相关。本课题组前期研究发现HO-1在氧化应激作用下发挥重要作用并成功建立可遗传性白内障小鼠模型,明确了钴原卟啉调控的HO-1与ARC相关性。本研究利用多聚精氨酸(R6)和钴原卟啉(CoPP)构建局部眼用的细胞穿透肽药物复合体R6-CoPP,通过体外实验对合成产物进行细胞毒性、细胞穿透性和抗氧化应激损伤能力的评估,通过体内动物模型实验初步评价其减轻晶状体上皮细胞氧化应激损伤的效应。研究发现:该复合体对细胞表现出的毒性较小,生物相容性较好,穿细胞膜能力强,预处理后的细胞受到氧化损伤时存活率明显上升。通过给予CoPP和R6-CoPP预处理后的硒性白内障损伤大鼠表现出晶体损伤减轻,晶体形态结构改善,抗氧化酶活性较损伤组增强。研究还证实在动物实验中,CoPP和R6-CoPP预处理可能通过上调晶体抗氧化酶活性来抑制晶状体上皮细胞凋亡,从而达到抗硒性白内障大鼠白内障生成和减缓其发展的保护作用,给未来的白内障预防和延缓其进展的药物研究提供一定理论和实验依据。
