Endometriosis is one of the most common gynecological diseases in women. The incidence rate of endometriosis is about 15%, the etiology is not clear, the rate of early diagnosis is low. Studies suggest that malignant transformation of ovarian endometriosis and ovarian clear cell carcinoma are closely linked, ovarian endometriosis is even considered to be precancerous lesion of the latter. But, the endometriosis carcinogenesis mechanism remains unclear. This study, intends to screen free protein of malignant transformation of endometriosis in blood by using the new serum-free primary cell culture system and mass spectrometry. Next, selected proteins were verified in blood and histological sample. This study will discuss the clinical value of selected proteins for screening high-risk groups of malignant transformation of ovarian endometriosis, predicting disease progression, classifing diseases and providing individualized treatment. The innovation of this research mainly in two aspects: the first, the patient who has a history of benign endometriosis and endometriosis-associated ovarian cancer is perfect for the object of study of endometriosis canceration mechanism; the second, the new serum-free primary cell culture system that of no extract serum, no interference of high abundance proteins will provide a guarantee for subsequent mass spectrometric analysis.
子宫内膜异位症是最常见的妇科疾病之一,在育龄妇女中发病率约为15%,发病原因不清,早期诊断率低。研究认为,内异症恶变与卵巢透明细胞癌存在密切联系,甚至被认为是后者的癌前病变,但目前对于子宫内膜异位症恶变机制仍不清楚。本研究中,拟采用新的无血清原代细胞培养体系,利用质谱分析技术,筛选子宫内膜异位症恶变的相关血液游离蛋白,并对筛选出的差异蛋白进行大样本量血液学和组织学层面的验证,探讨其临床应用价值,为临床筛选内异症恶变高危人群、预测疾病进展、进行疾病分型和个体化治疗提供重要基础理论依据。本研究的创新之处主要为两方面:一、既往有良性内异症手术标本的内异症相关性卵巢癌患者是研究子宫内膜异位症恶变发生机制极佳的研究对象,前期工作中已经留取到的标本珍贵;二、利用新的无血清原代细胞培养体系,该体系不包含垂体提取物和血清,即无高丰度蛋白的干扰,所有成分已知,为后续的质谱分析的准确性提供了保证。
