To meet the demand for high throughput and low operation cost in purification of recombinant human serum albumin(rHSA), continuous counter-current chromatography based on the principle of simulated motion of stationary phase was thoroughly studied with anion exchange media. Compared with batchwise chromatography in a fixed bed, the separation efficiency were remarkably enhanced in continuous counter-current chromatography. In the fundamental research, a mixture of human serum albumin and ovalbumin which exhibit very similar isoelectric points can be sucessfully separated using this technology. Also, with the continuous counter-current chromatography developed in the project, crude sample of recombinant human serum albumin with content of 23%-30% HSA can be purified to a high content of of 93%-97%. The research work in the project has provided a new practicable method for large-scale separation of proteins with low operation cost. The technology formed in this project has been applied for a Chinese invention patent.
为了满足重组人血清白蛋白分离提取规模很大而要求成本很低的需求,对其中的层析过程,提出了符合蛋白质层析特点的、依据固相吸附载体模拟移动原理构建的连续逆流蛋白质层析装置。通过提高载体的过程利用率明显节约载体用量,逆流接触提高传质推动力、减少流动相耗用量并提高浓度。使之成为一种新的高效经济的层析技术,并用于重组白蛋白的分离。.
