The item which revolved around the function of TGF-β1 and its receptors in the pathogenic mechanism of colon fibrosis in ulcerative colitis andeffection of TGF-β1 on the production of extracelluar matrix and the gene expression, study the mechanism of acupuncture and moxibustion treatment on colon fibrosis. .Experiment study on the basis of colon fibrosis in ulcerative colitis rats treated by acupuncture and moxibustion. Detecting the TGF-β1, TGF--βRⅠ, TGF--βRⅡ,Ⅰ,Ⅲ,Ⅳcollagens expressions in each group by using immune immunohistochemistry, and the TGF-β1, TGF--βRⅠ, TGF--βRⅡ,Ⅰ,Ⅲcollagens gene expressions by using RT-PCR and situ hybridization. Comparisoned by normal group, model group have remarkedly rised expressions of TGF-β1, TGF--βRⅠ, TGF--βRⅡand gene, but herbs-partitioned moxibustion treatment could significantly improve the abnormal status. Herbs-partitioned moxibustion, electr-acupuncture and SASP groups could notably decrease expressions of Ⅰcollagen mRNA, there were no marked difference among each group, but expressions of Ⅲ collagen mRNA were mostly decreased in the SASP group..Sixty-five patients in out-patient clinic were randomized into herbs-partitioned moxibustion group(32 cases) and bran-partitioned moxibustion group(33 cases), and other 30 cases treated by SASP be compared. observe The clinical effects, pathological change and the expressions of TGF-β1, TGF-βRⅠ, TGF-βRⅡ, Ⅰ, Ⅲ, Ⅳ collagen were measured before and after treatment by using hematoxylin and eosin stain, immune immunohistochemistry and RT-PCR. The results were recently fully recovery 17 cases, improvement 12 cases, inefficiency 3 cases in the herbs-partitioned moxibustion group and 11, 16 and 6 cases respectively in the bran- partitioned moxibustion group, 9, 11, and 10 cases in SASP group. Pathological observation manifested many pathologic characters such as chronic mucosal inflammation, mucosal neutrophilic granulocyte infiltration, goblet cell decrease, neutrophilic granulocyte infiltration between epithelial cells, crypt abscess, abscess beside crypt or mucosal ulcer, herbs-partitioned moxibustion treatment could effectually redress patients's mucosal lesion. Herbs-partitioned and bran-partitioned have markedly inhibited action to expressions of TGF-β1, TGF-βRⅠ, TGF-βRⅡ and the gene, could significantly reduce expressions ofⅠ, Ⅲ, Ⅳ collagen and Ⅰ, Ⅲcollagen genes, especially in the herbs-partitioned group. .The results hints that in the prevention and treatment of colon fibrosis in ulcerative colitis, moxibustion may reduce the expressions of TGF-β1, TGF-βRⅠ, TGF-βRⅡ and gene, and hence to block or inhibit the synthesis of Ⅰ, Ⅲ, Ⅳ collagens, and improve the structure and funtction of colon mucosa.
本项目围绕TGF--β1(TGF--β1与TGF--βRⅠmRNA)在溃疡性结肠炎肠纤维化发病机制中的作用及对细胞外基质的产生与其基因表达的影响,研究针灸抗肠纤维化的作用机制,以期证明隔药灸防治溃疡性结肠炎肠纤维化是通过抑制TGF--β1的产生和活性,从而减少或阻止细胞外基质的合成,为隔药灸治疗溃疡性结肠炎肠纤维化提供分子免疫学与形态学基础。
