It is the first time that we get formula of internal dosimetry in suspending culture cells and anchorage-dependent cells. To observe culture vessel smooth muscle, cell cycle, survival and apoptosis with different irradiation dosage; to determine the relationship between irradiation dose and effect and the best irradiation dosage point; to observe abnormal chromosome of smooth muscle cell and change of relative gene expression after irradiating by different molecular biological technics; to find out influence on smooth muscle cell proliferation, induction of apoptosis and potential key point; to compare the different irradiation sensitivity among rat smooth muscle cell, human umbilical vein endothelium and rat myocardium by using β ray. What we have done is to provide theoretical basis and effective therapy of preventing blood vessel restenosis with radionuclide in clinic. All the work have been done according to the original plan. We also research the effect of radionuclide on blood vessel endothelium.
观察不同剂量放射性核素照射后培养血管平滑肌细胞的增殖变化;确定辐射剂量与效应间的关系和最佳辐射剂量点;用各种分子生物学技术测量照射后的平滑肌细胞染色体异常和相关基因及其产物表达的改变;了解放射性核素照射对平滑肌细增殖的影响和对细胞凋亡的诱导作用及可能的关键环节;为临床应用核素预防在狭窄提供理论依据及有效的治疗方法。
