Twirling reinforcing-reducing manipulation is regarded as a critical factor of therapeutic effect , it also has long been a controversial focus of acupuncture and moxibustion cirle at home and abroad. Our research group has found that twirling reinforcing-reducing manipulation can regulate the blood pressure by activating the central target brain area, and there is a difference in the brain area of different acupuncture manipulation in the study of former National Natural Science Foundation of China. For further study on the central mechanism of the twirling reinforcing-reducing manipulation of acupuncture, we continue to choose the spontaneously hypertensive rats as the research object, selecting the key proteins and its pathways associated with the pathogenesis of hypertension. And then Western Blot and immunohistochemistry were used to validate and locate them. The purpose of this study is to reveal the central mechanism of twirling reinforcing-reducing manipulation from the perspective of protein molecules and provide new targets and theoretical basis for the prevention and treatment of hypertension, promoting the transformation of research results of hypertension to clinical application timely.
捻转补泻手法是实现针刺疗效的关键因素,也是长期以来国内外针灸界争论的焦点。本课题组在以往承担的国自然课题研究中发现,针刺捻转补泻手法可通过激活中枢靶脑区发挥其血压调控的作用,并且不同手法激活的脑区有差异。为进一步阐明针刺捻转补泻手法的中枢效应机制,本研究继以自发性高血压大鼠模型为研究对象,运用蛋白组学技术筛选靶脑区中的差异蛋白,选取与高血压发病机制密切联系的关键蛋白及其通路,进一步采用Western Blot、免疫组化技术对其进行验证和定位分析。以期从蛋白分子水平揭示捻转补泻手法的中枢机制,同时为高血压防治提供新的靶点和理论依据,促进高血压研究成果及时向临床转化。
