Clarifying the biosynthetic mechanism of antibiotics is an important task of the research on modern life sciences, and of great significance to make possible to regulate the microbial metabolism artificially, and then to increase the yield of antibiotic production from the microbial fermentation. In our former study, a new kind of nucleoside antibiotics with a new molecular structure was purified from the broth of the strain of Micromonospora carbonacea JXNU-1 with broad-spectrum antibacterial activity, and named the antibiotic JX temporarily, but it is not clear about the biosynthetic mechanism. Therefore in this study, the strain of Micromonospora carbonacea JXNU-1 will be used as material, the objective, firstly, is to study the possible precursor of the antibiotic JX by the "feeding test method" according to its structure, and the biosynthetic gene (cluster) and related proteins for the new nucleoside antibiotics JX production by genomics, transcriptomics and proteomics research methods respectively at the same time. Finally, to comprehensive analyse, deduce the biosynthetic mechanism of the new nucleoside antibiotics JX from Micromonospora carbonacea JXNU-1. The study may lay the foundation on the optimization of the process for the production, and then the enhanced yields of the new nucleoside antibiotics JX from Micromonospora carbonacea JXNU-1. It is of great theoretical and practical significance on further development of the new nucleoside antibiotic JX with autonomous intellectual property.
抗生素生物合成机制的阐明是现代生命科学研究的一项重要任务,对实现微生物代谢的人工调控、进而提高微生物抗生素的发酵产率具有重要意义。课题组前期研究获得的炭样小单孢菌JXNU-1所产抗菌单体组分经鉴定为一种新型核苷类抗生素,并被命名为抗生素JX。该抗生素具有广谱抗菌活性,但其自身生物合成机制仍不清楚。本研究以炭样小单孢菌JXNU-1为研究对象,根据抗生素JX的结构,采用"喂养试验法"研究抗生素JX生物合成的可能前体,同时采用基因组学、转录组学和蛋白质组学研究方法,研究抗生素JX生物合成的相关基因(簇)及相关蛋白(酶),在此基础上综合分析、推断炭样小单孢菌JXNU-1产新型核苷类抗生素JX的生物合成机制,为优化炭样小单孢菌JXNU-1产新型抗生素JX的过程、提高抗生素JX的产率奠定坚实的理论基础,对进一步研发具有我国自主知识产权的新型核苷类抗生素,具有重大的理论和实践意义。
