Tumor is a major disease serious harm to human health. Tumorigenesis can be led with lower body immunity following immune escape with occurrence of immunosuppression. Activation of immune monitoring, blocking the tumor on the body's immune suppression and reversal of tumor immune escape are the vital directions of therapy. PD-1/PD-L1 signaling pathway is the key channel for mediating tumor immune escape. Our previous works confirmed the improvement of cancer-related fatigue and lymphedema exposuring Zusanli(ST 36), Guan Yuan(RN 4) and other points with 10.6μm laser moxibustion. Therefore, this study is designed to observe the antitumor effect, the regulation of immune function and interpretation of PD-1/PD-L1 negative synergistic signaling pathway using 10.6μm laser moxibustion on the tumor-bearing mice inoculated with Lewis lung cancer cell, which is expected to reveal that laser moxibustion can play an active role in improving the body immunity and regulating the immune escape mechanism of tumor. The results will also provide scientific evidences for the antitumor effect and improvement of cancer related symptoms with moxibustion.
肿瘤是严重危害人类健康的常见重大疾病,机体免疫力低下可导致肿瘤发生,肿瘤发生后又会产生免疫抑制作用使肿瘤免疫逃逸。激活免疫监控,阻断肿瘤对机体的免疫抑制作用,逆转肿瘤免疫逃逸是重要的治疗方向。PD-1/PD-L1信号通路是介导肿瘤免疫逃逸的关键作用通道。本课题组前期研究发现,10.6μm激光灸照射足三里、关元等穴能改善癌症相关性疲劳和淋巴水肿。因此,本申请项目将通过观察10.6μm激光灸对Lewis肺癌株细胞接种的小鼠肿瘤的抑瘤效应、免疫功能调节及对PD-1/PD-L1负协同刺激信号通路的调控,阐释其作用机制,证实激光灸不仅能改善机体免疫功能,也可通过阻断PD-1/PD-L1负协同刺激信号通路,重新激活免疫系统,逆转肿瘤免疫逃逸。实验结果将为灸法临床抑制肿瘤,减少肿瘤复发和改善肿瘤相关症状提供科学依据。
机体免疫力低下可导致肿瘤发生,肿瘤发生后又会产生免疫抑制作用使肿瘤进一步逃逸免疫监控,其中,PD-1/PD-L1信号通路是介导肿瘤免疫逃逸的关键通道。本课题组前期研究发现,10.6μm激光灸照射穴位能改善癌症相关性疲劳和淋巴水肿。因此,本项目通过对Lewis肺癌株细胞接种的C57BL/6小鼠皮下肿瘤的体积测量、对免疫细胞及亚群分类的检测,对小鼠血清及脾脏、肿瘤组织内PD-1/PD-L1信号通路相关因子、蛋白的检测,探索10.6μm激光灸的抑瘤效应及作用机制。本研究结果显示:10.6μm激光灸连续照射足三里、关元15min/日,连续15d,对荷瘤小鼠肿瘤的生长具有一定的抑制作用,同时能减轻荷瘤小鼠肺部的肿瘤浸润。10.6μm激光灸可以调节小鼠外周免疫系统中Trg、NK细胞的比例,可以显著降低肿瘤组织局部M-MDSC细胞的比率(P<0.05)。10.6μm激光灸可降低血清中PD-L1、VEGF、CMTM6(P<0.05)的蛋白表达,减少肿瘤组织中PD-1、PD-L1(P<0.05)蛋白的表达,其机制可能与IFN-γ、IL-10有关。10.6μm激光灸与抗PD-1抗体联合使用对PD-1、PD-L1的抑制作用更加明显(P<0.05),对HIF-1a也有一定的抑制作用。证实了10.6μm激光灸不仅能通过调节免疫细胞改善机体免疫功能,也可通过抑制PD-1/PD-L1负协同刺激信号通路,重新激活免疫系统,减少肿瘤的免疫逃逸,从而发挥一定的抑瘤作用。研究结果将为肿瘤临床的联合免疫疗法提供相应的科学依据。本项目已发表直接相关论文3篇,其中SCI1篇;在投论文2篇,其中SCI1篇;参加2019国际肿瘤学术会议并做海报展示报告;共培养硕士研究生3名;项目组获得2021年上海市自然科学基金1项:HIF-α在艾灸抑制肺癌模型小鼠肿瘤微环境Tregs中的作用(21ZR1463300)。
