With the rapid development of GDP of our country, the production of nature rubber is less than 20% of the total consumption in our country. Rubber tree were planted in no-traditional cultivation area in China, and the development of rubber tree cultivation were restricted with cold stress. So it is important to develop cold tolerance breeding of rubber tree..In this study, wild germplasm with cold tolerance were screened by physiological characters, and the results of EST-SSR and physiological characters in the core collections were analyzed by association study, functional markers associated with cold tolerance were identified. Finally, the most prominent markers were analyzed by bioinformatical methods, and the genes closely related to cold tolerance were cloned. All of the results are useful to provide cold cross breeding and molecular breeding with new ideas and genes.
随着国民经济的不断发展,我国天然橡胶自给率已不足20%,天然橡胶种植业面临强大的增产压力,而我国属于非传统植胶区,低温寒害是制约我国橡胶树种植业发展的重要瓶颈,因此抗寒育种研究在橡胶树遗传育种中就显得尤为重要。本项目以橡胶树野生种质核心库为研究对象,通过抗寒指标测定从中筛选抗寒野生种质;将核心种质的EST-SSR分析结果与抗寒指标测定结果进行关联分析,从而开发出抗寒相关的EST-SSR功能标记;分析对抗寒性贡献率高的相关标记所在EST的信息,克隆与抗寒性关系密切的基因全长序列。项目的完成可以为我国橡胶树抗寒杂交育种和分子育种提供新思路和优异基因源,从而为抗寒育种奠定坚实的基础。
随着国民经济的不断发展,我国天然橡胶自给率已不足20%,天然橡胶种植业面临强大的增产压力,而我国属于非传统植胶区,低温寒害是制约我国橡胶树种植业发展的重要瓶颈,因此抗寒育种研究在橡胶树遗传育种中就显得尤为重要。本项目对441份橡胶树野生核心种质进行了抗寒性分析,利用脯氨酸含量、可溶性糖含量等生理指标与植株冻后恢复能力相结合的评价体系,筛选出Germ012、Germ017、Germ272等3份抗寒性较强的种质;利用115对EST-SSR引物对核心种质进行了基因型分析,通过对核心种质的抗寒性与基因型的关联分析,获得5个与抗寒性显著相关的SSR标记,贡献率在6.49%-10.22%。项目的完成可以为我国橡胶树抗寒杂交育种和分子育种提供新思路和优异基因源,从而为抗寒育种奠定坚实的基础。
