Adenomyosis has gradually become a common gynecological disease, difficult disease, and the incidence of its dysmenorrhea as high as 77.8 %. It seriously affects the quality of life of patients. The adenomyosis which with the dysmenorrhea is with the relevant uterine smooth muscle cells by ER and BKca, TRPV-1 ion channels in the abnormal control of excessive contraction. Our previous studies have shown that SGD can reduce the ERα mRNA of adenomyosis cells. So this project is to screen hypertrophy of uterine smooth muscle cells, in vitro cell culture experimental observation peony and licorice soup extracts the adenomyosis dysmenorrhea patients with lesions of proliferative activity intervention effect, using ELISA, immunocytochemistry, RT-PCR method to detect the expression of ER, of BKCa. Respectively, at the genetic level protein expression and other sectors to explore the Effect of the peony and licorice soup, seeks to clarify the deep mechanism of traditional Chinese medicine by the SGD, to provide experimental evidence and new research ideas.
子宫腺肌病已逐渐成为妇科常见病、疑难病,其痛经发生率高达77.8%,严重影响患者生存质量。子宫腺肌病痛经与子宫平滑肌细胞受ER与BKca、TRPV-1离子通道异常调控而过度收缩有关。我们的前期研究表明芍药甘草汤水提液可降低子宫腺肌病细胞ERαmRNA,故本项目拟通过建立子宫腺肌病痛经患者的病灶细胞体外体系,筛选其增生肥大的子宫平滑肌细胞,通过体外细胞培养实验观察芍药甘草汤水提液对子宫腺肌病痛经患者的病灶细胞增殖活性的干预作用,采用ELISA、免疫细胞化学、RT-PCR等法检测其对细胞ER、BKCa表达的调控,分别在基因水平、蛋白表达等环节探讨芍药甘草汤"一点多位"协同增效产生"多靶点"效应的深层作用机制,旨在阐明中药经方芍药甘草汤止痛的深层作用机理,为子宫腺肌病痛经的新药研发提供实验依据及新的研究思路。
项目背景:子宫腺肌病痛经严重影响患者生存质量。西医治疗手术创伤、西药副作用大、痛经易复发不被患者接受,而中医药在缓解疼痛、提高患者生存质量等方面有独特的优势,本课题在前期研究基础上进一步探讨芍药甘草汤为对其干预机制具有重要意义。.研究内容:取病理确诊为子宫腺肌病的病灶进行平滑肌细胞纯化培养,采用CCK-8、PCR、免疫细胞化学等方法检测芍药甘草汤及其单体对细胞增殖活性、E2、BKca α mRNA、TRPV1mRNA、ER、 BKca α、BKca β蛋白表达的影响。.结果:(1)CCK-8法测定芍药甘草汤对细胞增殖的抑制作用并进行药物浓度筛选,芍药甘草汤低浓度组(40mg/mL)对细胞的抑制率是40.08%,芍药甘草汤高浓度组(80mg/mL)对细胞的抑制率是58.06%。(2)与空白对照组相比,各组细胞E2自分泌均有不同程度的下降,芍药甘草高浓度组细胞E2表达差异有统计学意义(P < 0.05)(3)与空白对照组相比,芍药甘草高低浓度组、甘草苷高浓度组、芍药苷甘草苷高浓度组细胞TRPV-1 mRNA表达差异有统计学意义(P < 0.05);(4)与空白对照组相比,芍药甘草汤高低浓度组、芍药苷低浓度组细胞BKCa αmRNA表达均有不同程度的下降,差异均有统计学意义(P < 0.05);(5)与空白对照组相比,各组细胞ER蛋白表达均有不同程度的下降,芍药甘草高低浓度组、芍药苷高浓度组细胞ER表达差异有统计学意义(P < 0.05);(6)与空白对照组相比,芍药甘草汤高低浓度组、甘草苷高低浓度组细胞BKCa α蛋白表达均有不同程度的下降,差异均有统计学意义(P < 0.05);(7)与空白对照组相比,各组细胞BKCa β表达均有不同程度的下降,但差异均无统计学意义(P ˃ 0.05)。.科学意义:芍药甘草汤可能通过抑制子宫腺肌病病灶细胞增殖、降调子宫腺肌病病灶细胞ER、BKCa α、BKCa β蛋白表达及调控TRPV-1 mRNA、BKCa αmRNA基因水平,多靶点多角度效应调控,从而减缓了子宫平滑肌的收缩强度,有效缓解了患者的痛经症状,此可能是芍药甘草汤治疗子宫腺肌病的主要机理之一,为子宫腺肌病痛经的新药研发提供了实验依据。
