Recently, accumulating studies have suggested that glycogen synthase kinase-3β(GSK3β)and its related signaling pathways play critical role in both pathophysiology of depression and therapeutic action of antidepressants, and that induction of inhibitory serine-phosphorylation of GSK3 can promote antidepressant-like effect. Our previous work have demonstrated that the total flavonoids extracted from Xiaobuxin-Tang (XBXT-2) exerts excellent antidepressant-like effect in multiple animal models of depression, which may due to its up-regulation of hippocampal pGSK3β and phosphorylated cyclic-AMP response element binding protein (pCREB), reversal of serotonergic dysfunction and maintenance of hippocampal neuroplasticity. Upon these evidences and our previous results, in present study, we plan to investigate the effect of XBXT-2 on GSK3β and its related signaling pathways at whole animal level and cell level, with the application of multiple research approaches including behavioral pharmacology, cell and molecular technology, to determine whether GSK3β signaling pathway are causative mechanisms underlying the antidepressant-like effect of XBXT-2, which may provide a useful reference for antidepressant signaling pathway research of Traditional Chinese Medicine.
近年来研究发现糖原合成酶激酶3β(GSK3β)及其相关信号通路与抑郁症发生和药物抗抑郁作用密切相关,促进GSK3β磷酸化、抑制GSK3β功能具有抗抑郁作用。本研究前期发现小补心汤总黄酮(XBXT-2)具有明确的抗抑郁作用,可提高抑郁模型动物海马脑区GSK3β和cAMP 反应元件结合蛋白(CREB)磷酸化水平,并且具有增强5-HT神经传导功能和调节神经可塑性的作用。本项目拟在此基础上,应用行为药理学、细胞生物学和分子生物学技术等多种研究手段,从整体动物和细胞两个水平研究XBXT-2调控GSK3β及其相关信号通路的机制,以及该作用与其抗抑郁的关系,力争回答调控GSK3β信号通路是否是XBXT-2发挥抗抑郁作用的关键环节,同时也能为中药抗抑郁的信号通路机制研究提供有益借鉴。
该项目围绕GSK3β及其相关信号通路研究小补心汤总黄酮(XBXT-2)调控细胞内信号转导通路的抗抑郁作用机制,采用行为药理学和分子神经生物学相结合的研究手段,阐明了调控GSK3β信号通路是XBXT-2 发挥抗抑郁作用的关键环节,主要研究发现包括:(1) 采用多种抑郁动物模型,发现在XBXT-2发挥抗抑郁作用的同时,可促进抑郁模型动物海马GSK3β磷酸化,从而明确了促进GSK3β磷酸化是XBXT-2发挥抗抑郁作用的重要机制;(2)深入研究了XBXT-2调控调控GSK3β磷酸化的信号通路机制,明确了XBXT-2可同时激活PI3K信号通路和Wnt信号通路,而这两个通路的化学抑制剂均可阻断XBXT-2的抗抑郁作用;(3)AAV病毒介导的GSK3β(S9A)突变基因过表达可阻断XBXT-2的抗抑郁作用及其对信号通路的激活,进一步确认GSK3β是XBXT-2发挥抗抑郁作用的关键分子。该项目阐明了XBXT-2 抗抑郁作用的细胞内信号通路机制,并为其他抗抑郁中药的分子机制研究提供了有益借鉴。本研究成果发表SCI论文2篇、国内核心期刊论文3篇,获2014年度中华中医药学会科学技术奖一等奖和2015年度北京市科学技术奖一等奖各1项,培养毕业博士研究生1名、硕士研究生3名。作全国学术会议专题报告1次、墙报交流1次。
