Hypertension is an important public health problem, Qing Re Jie Du is one of the important Chinese medical treatment, and the decoction of Huang Lian Jie Du is a classic ancient formula. The applicants previously found that HLJDD had the effect of up-regulating the signals of eNOS/NO/ PKG and causing the over-expression of miR-133a, and the over-expression of miR-133a could cause the down-regulating endothelial cells’ Caveolin-1 and negative regulating the eNOS. In this project, we hypothesized that HLJDD up-regulating miR-133a decrease Caveolin-1 is probably the mechanism of up-regulating the content and the activity of eNOS. This project will treat the SHR with HLJDD and using Real-time Quantitative PCR and Western blot to detect the RNA and protein expression of mir-133a, caveolin-1,eNOS, and the activity of eNOS, and same experiments will do in vitro. This project will illuminate that the miR-133a/ Caveolin-1/eNOS path-way weather it involve the mechanism of the HLJDD up-regulating the eNOS/ NO signal of SHR aorta or not. The results will provide new evidences for the clinic usage of the ancient classic formula HLJDD.
高血压是重大公共卫生问题,清热解毒是中医防治高血压的重要治法之一,黄连解毒汤(HLJDD)是清热解毒的古典经方。申请者前期研究表明HLJDD对SHR主动脉的eNOS/NO/PKGⅠ信号上调、miR-133a过度表达,miR-133a高表达可使内皮细胞Caveolin-1下调,Caveolin-1 又负调控eNOS。推测HLJDD对SHR高表达的mi-133a使 Caveolin-1下调,并升高eNOS含量及活性。本项目通过HLJDD干预SHR,采用荧光定量PCR、Western blot检测主动脉mir-133a、caveolin-1、p-eNOS表达及eNOS活性,并以HLJDD含药血清干预牛主动脉内皮细胞,检测相应mRNA和蛋白表达,探讨miR-133a/Caveolin-1/eNOS通路在“HLJDD上调SHR主动脉eNOS/NO信号”作用机制,为清热解毒方临床应用提供实验支持。
高血压是最常见的心血管疾病,具有中医的“热”、“毒”特性,课题组前期研究表明黄连解毒汤(HLJDD)对自发性高血压大鼠(SHR)主动脉的eNOS/NO/PKGⅠ信号上调、miR-133a过度表达。因此本项目在既往研究基础上,以miR-133a/Caveolin-1/eNOS通路为研究切入点,从体内实验和体外实验开展研究,体内实验观察了HLJDD对SHR血压的影响,HLJDD对SHR血清NO、ET-1及主动脉eNOS含量变化的影响,采用分子生物学技术观察了HLJDD对SHR主动脉组miR-133a/Caveolin-1/eNOS通路相关基因和蛋白的表达;体外实验观察了HLJDD对牛主动脉内皮细胞(BAECs)上清液中NO、ET-1及eNOS含量变化的影响,并采用分子生物学技术观察了HLJDD含药血清对牛主动脉内皮细胞(BAECs)miR-133a/Caveolin-1/eNOS相关基因和蛋白的表达。结果表明HLJDD干预SHR一定时间后能够在一定程度上降低SHR血压且与量效有一定的关系,HLJDD能够在一定程度上升高血清中NO、eNOS含量,降低ET-1的含量达到保护血管内皮功能的作用,HLJDD可能通过升高SHR主动脉miR-133a、eNOS mRNA表达水平、升高eNOS、p-eNOS蛋白的表达水平、降低Caveolin-1 mRNA及蛋白表达水平达到保护SHR大鼠血管内皮功能,从而达到降低血压的作用;黄连解毒汤含药血清可能通过升高牛主动脉内皮细胞(BAECs)miR-133a、eNOS mRNA表达水平、升高eNOS、p-eNOS蛋白的表达水平、降低Caveolin-1 mRNA及蛋白表达水平发挥保护血管内皮功能的作用。本项目从动物、细胞、分子生物学实验,阐述了miR-133a/ Caveolin-1/eNOS 通路在“HLJDD 上调SHR 主动脉eNOS/NO 信号”中的作用机制,为中医药防治高血压提供了基础研究支持。
