Myocardial remodeling is a common precursor to heart failure in humans.The vitamin D deficiency and the imbalance of Th17/Treg mediated by dendritic cells persisted in the whole process of myocardial remodeling. Vitamin D plays a critical role in regulating the differentiation and maturation of dendritic cells. Dendritic cells are the specific professional antigen presenting cells (APC) play a key role in immune homeostasis which can directly activate the initial T cells into Treg cells or Th17 cells. Previous studies have found that icariin not only acts on the vitamin D axis interfere the myocardial remodeling,but also regulates the differentiation and maturation of dendritic cells. In this study ,we propose the mechanism of icariin is regulates the dendritic cells affect the balance of Th17/Treg by acting on the vitamin D system in treatment myocardial remodeling.We are about to reveal the immunological mechanisms of icariin intervention on myocardial remodeling and hope to provide a new target and experimental basis for traditional Chinese medicine treatment of cardiovascular disease.
心肌重塑是多种心脏疾病最终进展为心力衰竭的主要病理。维生素D缺乏及树突状细胞介导的Th17/Treg失衡持续存在于心肌重塑的发展过程。维生素D作用于维生素D受体调节树突状细胞免疫状态。树突状细胞是体内唯一能直接激活初始T细胞的最强专职抗原提呈细胞,能调节CD4+T向Th17细胞与Treg细胞分化,在免疫稳态中起着核心作用。研究表明,淫羊藿苷除能作用于维生素D轴,干预心肌细胞重塑外,还能调节树突状细胞免疫状态。据此提出:淫羊藿苷作用于维生素D轴调节树突状细胞功能,从而影响Th17/Treg平衡,干预心肌重塑的科学假说。课题拟选用淫羊藿苷干预Dahl盐敏感大鼠心肌重塑模型及模拟心力衰竭微环境血清下树突状细胞对CD4+T分化影响,结合细胞生物学、分子生物学研究方法,揭示淫羊藿苷干预心肌重塑的免疫学机制,为中医药治疗心血管疾病提供一个新的靶点及实验基础。
