Cancer stem cells (CSCs) play central role in the formation and development of cancer. Studies showed capsaicin has significant inhibitory effect on prostate cancer, however, the effects of capsaicin on prostate CSCs remain undefined. In our preliminary studies we found that capsaicin reduced the sphere formation of prostate cancer cells cultured in serum-free-medium, decreased CSCs markers’ expression, decreased PCAT-1 expression and suppressed Wnt/β-catenin pathway . We also found that PCAT-1 significantly increased in prostate CSCs. PCAT-1 may be a key oncogenic lncRNA in cancer, however, the role and regulation of PCAT-1 in prostate CSCs activity and capsaicin intervention has not been defined. Using serum-free-medium cultured prostate CSCs and nude mouse xenograft models, the proposed research will for the first time to investigate the involvement and function of PCAT-1/Wnt/β-catenin in prostate CSCs activity regulation. Moreover, this research will illustrate the role of PCAT-1/Wnt/β-catenin in the interventional effects of capsaicin on prostate CSCs. Findings from this research will provide important new insights into the molecular mechanisms of PCAT-1/Wnt/β-catenin in regulating prostate CSCs activity as well as its target intervention.
肿瘤干细胞对肿瘤发生发展具有决定性作用。研究表明,辣椒素对前列腺癌具有抑制作用,然辣椒素对前列腺癌干细胞的干预作用及机制尚不清楚。我们前期研究发现,辣椒素可显著抑制前列腺癌干细胞干性,下调PCAT-1表达,抑制Wnt/β-catenin通路活性;此外,我们发现PCAT-1在前列腺癌干细胞中高表达。研究表明,PCAT-1是重要致癌性lncRNA,且对Wnt/β-catenin通路具有调控作用,然PCAT-1是否调控前列腺癌干细胞干性及辣椒素的干预作用尚属未知。本课题拟采用无血清悬浮培养前列腺癌细胞模型和裸鼠移植瘤模型相结合,探讨PCAT-1/Wnt/β-catenin对前列腺癌干细胞干性的调控作用及机制,在此基础上研究辣椒素是否通过调控该途径活性对前列腺癌干细胞发挥干预作用。为揭示前列腺癌干细胞靶向干预的分子基础,阐明辣椒素对前列腺癌的干预作用及机制提供新的科学依据。
前列腺癌是男性最常见的恶性肿瘤之一。据全球肿瘤统计数据显示,其发病率和患病率居男性恶性肿瘤前列,已成为男性健康的重大威胁。肿瘤干细胞对肿瘤发生发展具有决定性作用。研究表明,辣椒素对肿瘤具有抑制作用,然辣椒素对前列腺癌干细胞的干预作用及机制尚不清楚。本课题采用无血清悬浮培养前列腺癌细胞模型和裸鼠移植瘤模型相结合,探讨辣椒素对前列腺癌干细胞的干预作用及PCAT-1/Wnt/β-catenin在其中的调控作用。本项目体内和体外实验结果均表明,辣椒素可显著下调CD133、CD44、ALDH1A1、OCT-4、Nanog和Sox2等前列腺癌干细胞分子标志物的表达,抑制前列腺癌干细胞的生长。进一步我们发现辣椒素可调控Wnt/β-catenin通路的活性,辣椒素可显著抑制该通路关键信号分子Wnt-2、p-GSK3β、β-catenin、c-myc和cyclinD1的表达,通过激活剂LiCl激活Wnt/β-catenin通路,辣椒素对前列腺癌干细胞的抑制作用显著减弱,表明Wnt/β-catenin通路在辣椒素干预前列腺癌干细胞干性中发挥重要调控作用。本项目亦发现辣椒素可显著抑制PCAT-1的表达,然转染PCAT-1低表达和过表达质粒均未发现其对前列腺癌干细胞干性具有显著的调控作用。同时,在本项目的研究过程中,我们发现无血清悬浮培养的前列腺干细胞球具有较强的EMT能力,而辣椒素可上调E-cadherin和ZO-1的表达,下调N-cadherin和Vimentin的表达,抑制前列腺癌干细胞EMT进程。上述研究结果可为揭示前列腺癌干细胞靶向干预的分子基础,阐明辣椒素对前列腺癌的干预作用及机制提供新的重要科学依据。
