Object: To study the binding of the SMN protein to the general transcription factor IIF in vitro. Method: Full length SMN protein, SMN truncated mutants D4, D3, D2 were expressed respectively using the GST fusion protein system. Pull-down assay in vitro and Western Blot were used to detect the binding of SMN protein and TFIIF. Results: Binding of the SMN protein and RAP30 was confirmed in vitro. Conclusion: These findings indicate an association of the SMN protein with TFIIF and suggest a role for the SMN protein in the RNA polII transcription/processing machinery.
