Peritoneal metastasis, as a major metastasis mechanism, predicts a miserable outcome, and anoikis resistance plays an important role in it. Our previous studies identified that PAK1 signaling pathway participates in anoikis resistance, CXCL12/CXCR4 signaling pathway involves in tumor associated macrophages (TAM) recruiting and N-acetylgalactosaminyltransferase family member 10 and 4 (GALNT10 and 4) play an essential role in the development of cancer. Further studies identified that GALNT10 expression is associated with miserable outcomes and peritoneal metastasis, and promotes the TAM infiltration and secretion of interleukin 6 in ovarian cancer. Based on the previous studies, we further identify the molecular mechanisms and functional activities of GALNT10 overexpression that recruites TAM and promotes anoikis resistance of cancer cells in ovarian cancer, and lay a foundation for the construct of molecular regulation and interference model to innovate a novel targeted therapy for ovarian cancer.
作为卵巢癌转移的主要途径,腹膜转移是患者预后不良的重要因素,失巢凋亡抵抗在其中发挥关键作用。项目申请人前期研究发现PAK1信号活化介导的失巢凋亡抵抗(Gastroenterolgy 2012)、参与肿瘤相关巨噬细胞招募活化的CXCL12/CXCR4信号(Nat Commun 2015)以及N-乙酰基半乳糖胺转移酶家族成员GALNT10(J Biol Chem 2015)和GALNT4(J Biol Chem 2017)在肿瘤发生发展中的关键作用。进一步研究发现卵巢癌患者肿瘤组织GALNT10高表达与患者术后生存期短和腹膜转移显著相关,其表达明显促进肿瘤相关巨噬细胞浸润并活化分泌IL-6。本申请项目拟在前期发现基础上,分析卵巢癌细胞GALNT10表达招募活化肿瘤相关巨噬细胞促进卵巢癌细胞抵抗失巢凋亡的分子机制及其参与腹膜转移的功能意义,建立相关分子调控和干预模型为开发针对性治疗药物奠定基础。
在本项目资助下,项目负责人围绕卵巢癌免疫逃逸和治疗抵抗展开研究工作,在肿瘤免疫微环境和靶向治疗诱导衰老方面取得了系列研究成果,以通讯作者在Cancer Immunology Immunotherapy, Cancer Chemotherapy and Pharmacology, Journal Gynecol Oncol等肿瘤学和妇产科学国际权威期刊发表7篇SCI 论文。我们发现卵巢高级别浆液性腺癌组织中GALNT10高表达患者预后不良,其肿瘤呈现免疫抑制微环境,其中发挥抗肿瘤效应的CD3 T细胞和NK细胞数量减少,发挥促进免疫逃逸的Treg细胞和M2巨噬细胞数量增加(Cancer Imnmuology Immunotherapy, 2020),该研究首次确认GALNT10与肿瘤免疫抑制微环境相关。PARP抑制剂奥拉帕尼靶向BRAC1/2突变卵巢癌患者,但获得性耐药影响了奥拉帕尼的疗效,研究发现奥拉帕尼小剂量长时间处理诱导卵巢癌细胞衰老相关表型,奥拉帕尼通过P53和P16依赖的方式诱导衰老相关表型(Journal of Gynecologic Oncology, 2019),而PPAR-γ抑制剂罗格列酮能逆转奥拉帕尼诱导的衰老相关表型,促进卵巢癌细胞凋亡,抑制肿瘤生长,同时罗格列酮联合奥拉帕尼抑制p53磷酸化和下调p21和p18蛋白水平(Cancer Chemotherapy and Pharmacology,2020),该研究为卵巢癌患者PARP靶向治疗提供了新的治疗策略。
