Irritable bowel syndrome (IBS) is an extremely common functional intestinal disease, which shown as abnormal gastrointestinal motility, sensation and secretary. The exactly pathophysiology of IBS is complex. Our previous studies have showed that abnormality of gastrointestinal hormone and intestinal gas in IBS patients may contribute to intestinal motility disorders. Recently researches have confirmed that the abnormality of short chain fatty acids(SCFAs) induced by abnormal colonic fermentation may contribute to intestinal motility disorders. In this project, the concentration of SCFAs in stool and serum of IBS patients and animal model will be detected.And the expression of SCFAs receptors and potassium channel ,calcium channel will be detected by western blot.The effects of serum in IBS patients ,SCFAs and three components on spontaneous contraction of colonic smooth muscle in each group are detected. In addition, the effects of serum in IBS patients ,SCFAs and three components on currents of ion channels on colonic smooth muscle cells are expected to detect by the whole-cell patch clamp. Meanwhile, the possible pathogenesis that abnormality of SCFAs in IBS patients may contribute to intestinal motility disorder are also about to be investigated, which may lay a theoretical foundation on developing specific curative medicine and method.
肠易激综合征(irritable bowel syndrome,IBS)是一种胃肠道运动,感觉及分泌异常的功能性胃肠病,其发病机制复杂。前期我们发现IBS患者体内胃肠激素及肠道气体异常参与肠道动力异常的发生,近期研究认为IBS患者结肠发酵异常导致短链脂肪酸(short chain fatty acids,SCFAs)含量异常也可能参与肠道动力异常。本项目检测IBS患者血清及大便SCFAs含量,利用westernblot检测IBS大鼠模型结肠SCFAs两种受体及钾通道、钙通道表达,采用IBS患者血清灌流大鼠结肠平滑肌检测肌张力变化,并采用IBS患者血清相应浓度的SCFAs及其各成分灌流检测肌张力,此外利用膜片钳技术检测IBS患者血清、SCFAs及其各成分灌流后大鼠结肠平滑肌细胞离子通道电流的变化,探讨SCFAs在IBS肠道动力异常中的作用,为寻找IBS的特异性治疗药物和方法提供理论依据。
肠易激综合征(irritable bowel syndrome,IBS)是一种胃肠道运动,感觉及分泌异常的功能性胃肠病,其发病机制复杂。近期研究认为IBS患者结肠发酵异常导致短链脂肪酸(short chain fatty acids,SCFAs)含量异常也可能参与肠道动力异常,因此本项目在动物模型、离体肌条及离体全结肠三个层面分别进行短链脂肪酸含量测定及肠道动力相关实验,明确短链脂肪酸在肠易激综合征中的含量及比例变化情况,初步探讨短链脂肪酸在肠易激综合征动力异常中的作用机制,实验主要侧重于短链脂肪酸对结肠平滑肌收缩幅度及收缩频率的影响和短链脂肪酸对全结肠运动模式的影响,并尝试应用短链脂肪酸对模型进行干预,为短链脂肪酸在临床中的应用和肠易激综合征患者低FODMAP饮食指导提供理论依据。
