本项目拟结合运用膜片箝记录、区段突变体扫描、定点突变、重组蛋白结合与磷酸化检测等技术,在原有工作基础上,深入研究酪氨酸激酶型受体EGFR、Trk、Ret等不同成员激活后对GIRK的调制作用,分析酪氨酸激酶型受体调制GIRK的特异性;确定酪氨酸激酶型受体调制GIRK的位点并明确其结构特点;分析GIRK的酪氨酸激酶型受体调制位点与已发现的G蛋白bg亚基、PIP2位点的关系;从而深入阐明酪氨酸激酶型受体调制钾通道GIRK的分子与结构基础。本研究对于认识酪氨酸激酶型受体在调制神经元兴奋性中的作用以及生长因子的调质样效应机制具有重要意义。
