The project aims to establish a rapid, accurate honey quality detection method based on the study of antioxidant activity in honey, providing theoretical basis for honey adulteration identification and geographical origins. In this study, a novel strategy for quantitative analysis based on chemometrics mathematical separation-enhanced HPLC–DAD (or HPLC–MS) is firstly developed, which applies chemometrics multi-way calibration algorithm, on account of the existing “second order or higher order advantage”, to realize simultaneous, direct and rapid, accurately qualitative and quantitative analysis of antioxidant activity components in honey. On account of the qualitative and quantitative results, we further establish the quantitative relation model between the antioxidants and honey antioxidant effect, illuminate the ratio between the antioxidant activity components, and study the microscopic mechanism of antioxidant effect in honey, and also estimate the performance of existing various antioxidant test method. In addition, on the basis of the qualitative and quantitative results of the antioxidant activity components, using chemometrics two-way analysis method, a clustering analysis is carried on for all the honey samples, in order to realize honey adulteration identification and geographical origins. The “quantitative advantage” of chemometrics multi-way calibration methods and the “qualitative advantage” of two-way analysis methods complement each other in this project, and the effective combination of these two methods has not been reported previously. This method could provide a powerful reference to fight with honey adulteration,confusion of botanical origin as well as honey of inferior quality.
本项目旨在基于化学计量学方法进行蜂蜜抗氧化活性研究,建立一种快速、准确的蜂蜜品质控制方法,为蜂蜜掺假鉴别和产地溯源提供理论研究基础。基于化学计量学“数学分离”辅助HPLC-DAD(或HPLC-MS)的绿色分析方法,利用存在的“二阶或高阶优势”,在蜂蜜复杂背景与干扰共存下实现多种抗氧化活性组分同时、直接、快速、准确定量分析。基于定性定量结果,建立抗氧化活性组分与蜂蜜抗氧化能力的定量关系模型,明确蜂蜜中抗氧化活性组分之间的比例关系,研究蜂蜜呈现抗氧化作用的微观机理,判断现有各种抗氧化测试方法的优劣。在此基础上,结合化学计量学一阶校正方法对定量结果进行聚类分析,实现蜂蜜的掺假鉴别和产地溯源。这种将化学计量学多元校正方法的“定量优势”和一阶校正方法的“定性优势”取长补短、有效组合的研究思路尚未见报道。本项目预期研究成果可以为解决目前蜂蜜市场上品种混乱、品质不佳、掺假造假等问题提供一定的参考和指导。
蜂蜜是天然抗氧化物质,具有很高的营养价值受到越来越多消费者的青睐。蜂蜜掺假形式多样,检测手段单一,具有一定的局限性和不确定性。本项目基于化学计量学多元校正方法的“定量优势”和一阶校正方法的“定性优势”,进行了蜂蜜抗氧化活性研究,并在此基础上,系统研究了蜂蜜品质控制的新方法。首先,利用化学计量学“高阶优势”,研究了高维数据的处理方法。通过对比二阶校正方法和三阶校正方法对复杂数据的分辨能力,深入挖掘和探讨了“三阶优势”。基于三种常用化学计量学二阶校正方法,对色谱的新型分析策略进行了探讨,研究了处理色谱峰重叠、色谱漂移等复杂数据时的表现性能等。其次,利用化学计量学方法,建立了蜂蜜抗氧化活性组分定性定量分析新方法,明确了蜂蜜抗氧化活性组分的种类和准确含量。基于多种抗氧化活性组分的定性定量结果,利用化学计量学一阶校正方法进行了聚类分析,建立了蜂蜜活性物质的定量组成与抗氧化活性之间的关系模型,鉴别出了关键抗氧化活性组分,实现了不同蜜源蜂蜜和不同掺假蜂蜜的品质鉴别。研究结果可为解决目前蜂蜜市场上品种混乱、掺假造假等现象提供参考和指导,还可以为蜂蜜加工产品、农产品、中药等物类的精准识别和分类研究提供参考,具有一定的理论价值和良好的应用前景。
