It is an urgent problem to improve the curative effect of gastric cancer. Immunodetection point PD-1 / PD-L1 inhibitors have become an important treatment for malignant tumors, which was also approved by FDA in the clinical application of gastric cancer last year. On the basis of the latest found that the “exhaustion of T cell”was associated with methylation of the major immune response factors after a period of PD-1 application, immune response woud be regain “activation” by the application of demethylation drug 5-Aza-dC. Therefore, we connected anti-PD-1 antibody on the biocompatible polymer material of to PEG-PCL and loaded 5-Aza-dC. By establishing a subcutaneous tumor of gastric cancer and in situ tumor model to simulate the biological characteristics of human tumors, the effect of the drug-loaded system on the targeting of PD-1+CTL cells and the methylation level of major immune response factors, the antitumor efficiency and its possible molecular mechanism will be explored. We are hoping the project will provide a scientific basis for the clinical application.
提高胃癌临床治疗疗效,是目前迫切需要解决的问题。免疫检查点PD-1/PD-L1抑制剂已成为恶性肿瘤重要的治疗手段,去年被FDA批准于胃癌临床应用。本项目申请人在前期工作成功掌握抗体连接于聚乙二醇-聚己内脂(PEG-PCL)二亲嵌段共聚物(可生物可降解的高分子材料)的基础上,以最新的研究发现PD-1单抗应用一段时间后再次出现“T细胞耗竭”与主要免疫应答因子发生甲基化有关,去甲基化药物5-Aza-dC可再次“活化”免疫应答为依据,选用PD-1单抗共轭连接于PEG-PCL纳米载体,构建负载5-Aza-dC的PD-1靶向纳米粒子。通过建立胃癌的皮下移植瘤及原位瘤模型来模拟人体肿瘤的生物学特性,研究该纳米载药体系对PD-1+CTL细胞靶向作用及主要免疫应答因子甲基化水平的影响,进一步探讨其对肿瘤的抑制作用及可能的分子机制,为靶向载体药物向临床应用过渡提供科学依据。
