Invasion and metastasis is the main cause of treatment failure in patients with colorectal cancer(CRC), Clarifying the molecular mechanisms of invasion and metastasis has important theoretical and practical significance in the prevention, diagnosis and treatment of CRC. In our previous studies, we found that flotillin-2 (FLOT2)was overexpressed in CRC cells and tissues, which may be closely correlated with the invasion and metastasis of CRC. Based on our previous studies,our research group will try to further elucidate the relationship between FLOT2 expression status and the clinicopathologic parameters and prognosis of CRC on larger samples; gene transfection and RNA interference (RNAi) techniques will also be applied to observe the effects of over- or low-expression of FLOT2 on the malignant biological behavior of CRC cell; establish whole-body visualizing animal model will be established and dynamicly observe the process of CRC formation and metastasis could be dynamicly observed; the effects and mechanisms of Cdc42-N-WASP-Arp2/3 pathway when FLOT2 regulates the invasion and metastasis of CRC will also be investigated. This study aims to expound the biological functions and the regulation mechanisms of FLOT2 in the invasion and metastasis of CRC, and provide theoretical basis for the further research based on the diagnosis、treatment and molecular targeted drug of FLOT2.
侵袭和转移是导致结直肠癌患者治疗失败的主要原因,明确结直肠癌侵袭和转移的分子机制对结直肠癌的预防、诊断和治疗具有重要的理论和实际意义。本课题组前期研究提示FLOT2在结直肠癌细胞和组织中高表达,而且可能与结直肠癌的侵袭和转移有关。我们拟在前期研究基础上,扩大临床样本数量,明确FLOT2表达状态与临床病理学参数、患者预后的关系;利用基因转染和RNAi技术,构建FLOT2稳定高/低表达细胞株,研究其对结直肠癌细胞恶性生物学行为的影响;建立整体可视化人结直肠癌裸鼠皮下成瘤及转移瘤动物模型,动态对比观察肿瘤细胞的成瘤及转移情况;研究Cdc42-N-WASP-Arp2/3通路在FLOT2调控结直肠癌侵袭和转移过程中的作用及其机制。本研究旨在阐明结直肠癌中FLOT2表达上调的生物学功能和调控机理,为进一步研究基于FLOT2的诊断、治疗和靶向药物开发奠定理论基础。
侵袭和转移是导致结直肠癌患者治疗失败的主要原因,明确结直肠癌侵袭和转移的分子机制对结直肠癌的预防、诊断和治疗具有重要的理论和实际意义。运用RT-PCR、Western blot和IHC技术检测证实FLOT2在结直肠癌组织中呈高表达。分析FLOT2表达与结直肠癌患者临床病理特征及预后的关系,提示FLOT2的表达与结直肠癌的临床分期、侵袭和转移相关,FLOT2表达可作为预测结直肠癌患者不良预后的独立指标。利用基因转染和RNAi技术,构建FLOT2稳定高/低表达结直肠癌细胞株,采用transwell、划痕实验研究FLOT2对结直肠癌细胞恶性生物学行为的影响。划痕实验表明FLOT2增强了结直肠癌细胞的转移能力。Transwell迁移实验和基质胶侵袭实验验证FLOT2增强了结直肠癌细胞的侵袭及迁移能力。建立结直肠癌裸鼠转移瘤动物模型,体内观察证实FLOT2促进了结直肠癌细胞的侵袭转移。显微镜下观察发现,结直肠癌细胞内FLOT2 表达的变化,会引起结直肠癌细胞伪足的变化。利用激光共聚焦显微镜,证实FLOT2 和 Cdc42 蛋白共定位于结直肠癌细胞浆内。运用Western blot技术,证实在结直肠癌细胞中,FLOT2通过FLOT2-Cdc42-N-WASP-Arp2/3 通路来调节伪足的变化,从而发挥生物学功能。本研究初步阐明结直肠癌中FLOT2表达上调的生物学功能和调控机理,为进一步研究基于FLOT2的诊断、治疗和靶向药物开发奠定理论基础。在本项目研究过程中,培养了3个研究生,1篇SCI已可以在PUBMED中检索,1篇SCI已接收,1篇SCI已完成撰写,1篇中文文章已发表。
