It has been reported that hypoxia inducible factor-1α (HIF-1α) is involved in the pathological process in acute lung injury. The modulation of HIF-1α in the lung inflammation after acute lung injury has been proven. However, the role of HIF-1α in the vascular repair after acute lung injury remains unclear. Our preliminary data shows that HIF-1α regulates the vascular repair following lung injury. In this project, employing endothelial cell-restricted HIF-1α knockout mice, human lung microvescular endothelial cells, HIF-1α inhibitor and siRNA, we aim to identify whether endothelial cell-specific HIF-1α modulates the vascular repair following acute lung injury induced by lipopolysaccharide (LPS) and the involved molecular mechanisms. The results will develop the understanding of modulation of vascular repair following acute lung injury and identify the involved molecules in the regulation. In addition, the project will provide the potential therapeutic approaches for the treatment of acute lung injury.
低氧诱导因子1α(HIF-1α)在急性肺损伤的病理过程中具有重要的调节作用,其在肺急性炎症过程中的作用已经得到初步证实,但HIF-1α在肺血管损伤后修复过程中的作用尚不明确。前期研究提示HIF-1α可能参与急性肺损伤后血管修复过程。本项目拟以血管内皮细胞特异性HIF-1α敲除小鼠、人肺微血管内皮细胞为模型,以脂多糖刺激为处理手段,以HIF-1α抑制剂和siRNA作为工具,阐明血管内皮细胞HIF-1α是否参与急性肺血管损伤后修复过程,初步确认HIF-1α在肺血管损伤后修复过程中的作用及其机制。本项目将深化对急性肺损伤后血管修复过程调节的理解,寻找参与调节的分子,还将为研制肺损伤保护药物提供依据。
研究血管内皮细胞低氧诱导因子-1a(Hypoxia inducible factor-1a,HIF-1a)对急性肺损伤后血管修复过程的调控,及其该过程的信号通路,继而阐明HIF-1a介导急性肺损伤后血管修复的作用,有助于加深肺损伤后血管修复过程的理解,对寻找新的肺损伤保护药物靶点也具有重要意义。本项目拟以培养肺血管内皮细胞和血管内皮细胞特异性HIF-1a缺失小鼠为工具,研究脂多糖刺激和/或盲肠穿孔手术模型致败血症后,肺血管损伤后修复过程中HIF-1a的功能。.体外培养血管内皮细胞实验表明,脂多糖刺激可导致内皮细胞HIF-1a激活,诱发细胞炎症反应,导致细胞缝隙连接破坏,跨细胞电阻降低,提示细胞屏障功能减退。在小鼠实验中,脂多糖刺激或盲肠穿孔手术模型均可导致野生型小鼠肺血管出现损伤,内皮细胞出现凋亡,血管通透性增加,组织HIF-1a表达先上调后下降;恢复期过程中,肺血管内皮细胞出现增殖,损伤逐渐恢复。而在内皮细胞HIF-1a缺失小鼠中,肺血管损伤程度与野生型小鼠相同,但血管修复功能减退,血管内皮细胞增殖受阻,提示血管内皮细胞HIF-1a可调节肺损伤后血管修复过程。通过脂质体介导血管内皮细胞基因表达,损伤修复期血管内皮细胞HIF-1a高表达后,内皮细胞HIF-1a缺失小鼠血管修复功能回复正常。进一步的研究表明,肺血管内皮细胞HIF-1a表达可诱导转录因子FoxM1的表达,继而引起血管内皮细胞增殖,血管开始修复;而HIF-1a缺失后,FoxM1表达降低,血管修复功能减退。本项目证明了血管内皮细胞HIF-1a和FoxM1参与了肺损伤后血管修复过程。此外,还研究了HIF-1a参与了神经细胞凋亡和增殖的过程,扩展了对HIF-1a在细胞损伤病理过程中作用的理解。本项目已发表了8篇论文(其中2篇SCI论文),扩展了对于HIF-1a功能的认识。
