leukemia stem cells have the crucial roles in leukemia initiation and progression. It is the new strategy for the treatment of leukemia to find a kind of natural drug to kill the LSCs and protect the normal hematopoietic function. Our previous studies show that ginsenoside Rg1 has bidirectional effects of hematopoietic stem and progenitor cells ageing alleviation and K562 cells ageing promotion. It is worth studying the effect of Rg1 induces the leukemia stem cell senescence to treat leukemia. The lasted study indicated that SIRT1-mTOR mediated autophagy signaling pathway has the crucial role in cell senescence. It has not seen report about the effect and mechanism of SIRT1-mTOR signaling pathway in treatment of leukemia by Rg1 from inducing leukemia stem cell senescence. In this study, it takes LSCs as the target, in vivo and in vitro, using the latest techniques of stem cell and morphological and molecular biology methods to investigate the effect of SIRT1-mTOR pathway in LSCs senescence induced by Rg1. It can elucidate the effect and mechanism of Rg1 to cure leukemia in stem cell level. The study aimed to provide the evidence of the effective target of natural drug component promote tumor cell senescence, significantly, it will provide original approach in clinical leukemia therapy.
白血病干细胞(LSCs)在白血病发生发展中起决定作用,寻找能靶向清除LSCs,又能保护正常造血功能的天然诱导剂成为治疗白血病研究的新策略。我们研究显示:人参皂苷Rg1具有延缓造血干/祖细胞衰老和促进K562细胞衰老的“扶正祛邪”双向调控作用。Rg1能否通过诱导LSCs衰老发挥治疗白血病的作用值得深入研究。最新研究报道SIRT1-mTOR介导的自噬通路在细胞衰老过程中发挥重要作用,迄今SIRT1-mTOR通路在Rg1诱导LSCs衰老发挥治疗白血病中的作用机制还未见报道。本研究以LSCs为作用靶点,采用干细胞最新研究技术和形态学、分子生物学等方法,从体内和体外两条途径探讨SIRT1-mTOR通路在Rg1诱导LSCs衰老中的作用,阐释补气要药人参在干细胞水平上发挥“驱邪”治疗白血病的作用及机制。本项目旨在为天然药物有效成分促进肿瘤细胞衰老的研究提供实验依据,为临床治疗白血病提供新思路和新方法。
