The total of 1200 base pair sequences of exon 2,3,4,and intron 2 and 3 of Glucose-6-phosphate dehydrogenase gene from 15 insect species were amplified using nested PCR technique and then directly sequenced via BIGDYE sequencing kits. The primary analysis results show that both exons and introns of the Glucose-6-phosphate dehydrogenase gene are extremely conserved during insect evolution. The phylogenetic relationships of tested insects were constructed through analyses of DNA sequences and their translated protein sequences. This is the first study on dehydrogenase gene family from comparative genomics perspectives, and the data provide important clues to understand gene structure organization and sequence evolution of dehydrogenase gene family. Beside sequencing and analysis of Glucose-6-phosphate dehydrogenase gene, we also conducted the following studies:.(1).The origins and evolution of insect-specific genes and gene families using comparative genomics and bioinformatics tools.(2)We also conducted comparative mitochondrial genomics using long-distance PCR technique amplifying whole mitochondrial genome.(3)We also applied the Expert System methods to the taxonogy of China Acridoidea, and designed the new software -- Expert System of Classification, Retrieval and Diagnosis for China Acridoidea (ESCA), as an effective tool of Classification Retrieval and Diagnosis in our country's manage and survey of the grasshopper disaster.
