The mortality rate of Xinjiang Kazak esophageal cancer is higher than the average level in China. It is one of the malignant tumor which need focal prevention and cure. Non coding RNA is involved in a variety of important signal transcription processes, and plays an important role in the regulation of embryonic development and disease. We selected six cases of esophageal squamous cell carcinoma and adjacent tissues by microarray chip for the high-throughput analysis of circRNA and mRNA expression profile. We selected circRNA-MMP1 as the test subjects. In this study, we proposed to use the clinical specimens and esophageal squamous cancer cells as research materials. We carry out the research as follows: (1) analysis of the relationship between circRNA-MMP1 and clinicopathological features of esophageal squamous cell carcinoma patients; (2) analysis of the relationship between circRNA-MMP1/miRNA-145/MMP1 by bioinformatics; (3) analysis of the biological function of circRNA-MMP1 by over-expression and knock-down the circRNA-MMP1 expression. Through the research, we hope to find the effective target for the diagnosis and treatment of esophageal squamous cell carcinoma. To provide powerful theoretical and experimental basis for its application.
新疆哈萨克族是食管鳞癌的高发民族且死亡率远高于我国平均水平,是本地区重点防治的恶性肿瘤之一。非编码RNA参与多种重要的信号转导调控过程,在胚胎发育及疾病发生等过程中起重要调控作用。我们前期用微阵列芯片对6例食管鳞癌及癌旁组织进行circRNA、mRNA差异筛选,选择circRNA-MMP1为后续研究对象。本研究拟利用临床标本和体外培养的食管鳞癌细胞,以circRNA为切入点,开展如下研究:(1)分析circRNA-MMP1与食管鳞癌临床病理特征、患者预后的相关性;(2)利用生物信息学分析circRNA-MMP1/miRNA-145/MMP1相互关系;(3)利用功能获得与缺失实验,在食管鳞癌细胞和裸鼠移植瘤模型中研究circRNA-MMP1在食管鳞癌发生、发展过程中的生物学功能。以期通过以上研究找到有效的食管鳞癌诊断、基因治疗靶点,为临床食管鳞癌的诊断、治疗及预后提供有力的理论和实验依据。
新疆哈萨克族是食管鳞癌的高发民族且死亡率远高于我国平均水平,是本地区重点防治的恶性肿瘤之一。非编码RNA参与多种重要的信号转导调控过程,在胚胎发育及疾病发生等过程中起重要调控作用。在过去的十几年间,针对miRNA的功能与作用机制已被广泛研究,相比较而言,circRNA的潜在功能及其生物学特点仍不明确,其很可能与各种类型肿瘤的发生、发展有密切关系。随着高通量测序及生物信息学技术的迅猛发展,越来越多的circRNA被发现,其功能也在不断被揭示。我们前期用微阵列芯片对6例食管鳞癌及癌旁组织进行circRNA、mRNA差异筛选,选择circRNA-MMP1为后续研究对象。本研究利用临床标本和体外培养的食管鳞癌细胞,以circRNA为切入点,开展如下研究:(1)利用80对食管癌及癌旁组织样本进行定量检测后,并分析其与临床病理特征、预后等数据的相关性。在进行组织样本的qRT-PCR验证的同时,也检测了食管癌细胞株Eca109、TE-13、KYSE140和OE33细胞的LOC440173、hsa_circ_0025580和hsa_circ_0024108表达的情况。为了进一步进行后续研究我们选择Eca109和TE13作为研究对象。(2)对前期芯片数据进行深入的生物信息学分析,并利用组织样本进行数据的验证。(3)利用生物信息学分析circRNA-MMP1/miRNA-145/MMP1相互关系。以期通过以上研究找到有效的食管鳞癌诊断、基因治疗靶点,为临床食管鳞癌的诊断、治疗及预后提供有力的理论和实验依据。
