The extracts of forty edible mushrooms and plants were screened in vitro. Finally, twelve samples had strongly free radical scavenging and antioxidantive activities. F21 plant among twelve samples exhibited 90% superoxide scavenging activity, 79% hydroxyl radical scavenging activity, 74% inhibiting erythrocyte hemolysis and 86% inhibiting lipid peroxidation in rat brain homogenates. Therefore, F21 was further studied. By CM-cellulose, three unabsorbed peaks were collected. F21-3 had the highest potency in antioxidative activity. F21-3 is a small molecular. Spectroscopies studies using NMR and FTIR revealed F21-3 structure and inferred the relationship of structure and effects. Sometimes, the antioxidantive mechanism of F21 was inferred at tissue, cell and molecular levels. The results showed that F21 increased the SOD gene expression level in vivo, thus exhibiting the high antioxidantive activity. Therefore, the researches lay down the theoretical foundation for the application of F21
以食用药用菇类或植物中,选筛具有自由基清除和抗氧化作用的生物活性物质。并对其作用机制从分子水平上进行探讨,从而使生物活性物质作用机制的研究提高到一个新的水平。同时利用快速老化的小鼠模型,直接进行活性物质的抗衰老实验,以阐明自由基和抗氧化作用与衰老的关系。最终确定一种高自由基清除活性和抗氧化作用的制剂,即抗衰老保健品。
