用免疫组化、组化电镜和脑片原位杂交方法研究斜视性弱视发生发展过程中NMDA受体在视中枢细胞、超微结构和mRNA水平的变化。发现,1、斜视幼猫视皮层NMDA-R1的表达在斜视术后第一周开始减少。2、斜视猎视皮层细胞膜上NMDA-R1受体的突触数目,斜视术后第三周开始减少。3、NMDA-R1 mRNA R1-a,R1-b,R1-1构型在斜视猫视皮层的表达减少主要在IV层;R1-3构型的表达比正常猫增加,主要在第V-VI层。4、外膝体和视皮层斜视性弱视猫Y-细胞系统的Cat-301阳性细胞数比正常猫减少。结论:1、NMDA与视觉发育可塑性机制有关,参与形成了弱视视力功能损害的结构基础。2、斜视导致视皮层神经元NMDA受体水平表达减少源于其mRNA特异构型的转录抑制。3、Y-系统神经元表面抗原在弱视动物明显丢失,提示异常视觉环境可使外膝体和视皮层特异细胞群在分子水平发生改变。
