β-carotene is one of the most important carotenoids with the highest vitamin A activity and antioxidant activity. However, poor storage stability and low bioavailability strictly restricted its application in food industry. Therefore, the project firstly plans to prepare carboxylmethyl starch with high degree of substitution by noncrystalline and carboxymethylation treatments. A stable, efficient, and good biological compatibility of β-carotene nano-emulsion in starch-based microgel system was obtained by cross-linking carboxylmethyl starch via Fe3+ ions by oil in water in oil double emulsions method. The formation mechanism of this system and its improving the stability and bioaccessibility of β-carotene were investigated. The theory of β-carotene being targeted release in simulated intestinal fluid will be interpreted. Based on layer-by-layer self-assembly technique, β-carotene is controlled release by regulating and controlling the number of layers and amount of the positive and negative polyelectrolyte. These studies can provide some scientific basis for preparing β-carotene gut targeted delivery system and promoting the theory in controlled release system.
β-胡萝卜素是一种具有高维生素A原活性和强抗氧化特性的类胡萝卜素,其天然形式存在储藏稳定性差及生物利用率低等问题,限制其在食品工业中的广泛应用。基于此,项目拟从研究淀粉非晶化和羧甲基化制备高取代度羧甲基淀粉入手,基于Fe3+离子具有螯合羧基基团的性质,采用油包水包油双乳液法,构建稳定、高效、生物兼容性强的淀粉基微凝胶包β-胡萝卜素纳米乳液体系。探索体系形成、提高β-胡萝卜素储藏稳定性和生物利用率的机理;阐明体系中β-胡萝卜素在肠液内靶向释放原理;基于层层自组装技术,调控正负聚电解质的层数及包裹量等关键因子,揭示层层自组装稳定的体系中β-胡萝卜素在肠液内控释规律。通过本项目的研究,为β-胡萝卜素肠道靶向控释体系新途径、新方法的构建及控释理论的创新提供科学依据。
β-胡萝卜素是一种具有高维生素A原活性和强抗氧化特性的类胡萝卜素,其天然形式存在储藏稳定性差及生物利用率低等问题,限制其在食品工业中的广泛应用。本项目基于醇限制淀粉膨胀特性,采用不同醇热处理玉米淀粉,制备出非晶颗粒态淀粉及非晶颗粒态羧甲基淀粉。通过化学交联的方法制备了不同取代度羧甲基淀粉微凝胶和不同交联度羧甲基淀粉微凝胶,该微凝胶具有pH和离子强度响应性。研究微凝胶对蛋白的包埋特性,构建微凝胶/β-胡萝卜素纳米乳液载体系统,并阐明该系统中β-胡萝卜素在肠液内靶向释放原理。基于层层自组装技术,通过调控壳聚糖和羧甲基淀粉的层数和包裹量,体外模拟胃肠液的释放结果证明:两层包裹微凝胶客体复合物能有效的实现客体物质在胃液中释放较少,并进一步揭示其肠道缓慢靶向释放机理。该项目目前以第一/通讯作者发表标注SCI论文13篇,影响因子>5的3篇,其中ESI高被引论文4篇,封面论文1篇,包括Journal of Agricultural and Food Chemistry, Food Hydrocolloids和Food Chemistry等。申请国家发明专利6件。
