The analysis for the single and segment chromosome is a new cytogenesis technique, based on molecular method and molecular technique. The platform is already established for the study the chromosome of mode crop. Now, we introduced this technique into the analysis for the chromosome of woody plant. Because the study of tree chromosome is more difficulty than one of other plant, herbage or crop, and the result often is not stabilized, the study of tree chromosome is rare. With a micro glass needle under the reverted microscope, the singe chromosome on Cunninghamia lanceolata were microisolation, and Chromosome 4 was selected. Through the degenerate olionucleotide- primed- PCR amplification ( DOP-PCR ), DNA product were tested with DNA and Southern Test. The result shows that PCR product is from Chromosome 4 and proved that the Chromosome analysis based on molecular technique is feasible. With this technique, the special molecular markers (RAPD) on chromosome 4 were founded. We used same method to analysis other chromosome and a lot of special molecular markers also were founded. With those markers , the new chromosome coding is worked on. Meanwhile, some of special markets are transferred into Scar markers. To sum up, it is necessary to establishment of new analysis of chromosome for woody plant. It is tool to enhance the tree chromosome study, and bridge to connect the cytogenesis study on model plant.

采用显微操作技术,对杉木染色体或重要片段进行微分离与微切割,并进行体外扩增,建立染色体或特定片段的DNA文库,筛选和确定杉木染色体或重要片段的特异标记探针。使得目前部分遗传研究从林木的整个染色体组水平缩小到一个染色体,甚至是某个重要染色体的主要片段上,为开展林木改良有关的重要基因定位或染色体的片段上的测序工作奠定基础。.
