This project intends to adopt PCR, fingerprint analysis, immunohistochemical method, evaluation of intestinal flora differences in EV infection type and influence in the process of severe occurrence mechanism, confirmed that the intestinal flora differences apparent characteristics, can be obtained through the analysis of the constitution on discerniblile syndrome of traditional Chinese medicine clinical apparent objective evidence to predict enterovirus heavy vulnerable groups, so as to improve clinical disease prognosis and proprietary Chinese medicine curative effect into full play.
肠道病毒严重危胁儿童生命与健康,目前疫苗推广有效预防部分类型病毒感染(如IPV、HAV),但病毒变异后出现耐药性不可抗拒,从根本上发现重型易感人群特征具有重要意义。肠道病毒71型可引起重症手足口病,属中医“痘疹”范畴,《医宗金鉴·痘疹》中有关“禀赋顺逆”论述提示我们不同体质儿童感邪后疾病转归预后不同,前期研究及预试验发现气候、血清型、腹泻症状、气血盛衰影响EV感染类型及重症发生趋势,不同体质人群肠道菌群存在差异。由此我们提出不同体质儿童肠道菌群改变介导RIG-1信号通路调节SCARB2表达从而影响EV感染类型及重症发生趋势。本项目拟采用16SrRNA PCR、DGGE、ELISA、免疫组化,评价肠道菌群差异在EV感染、免疫反应及重症发生过程中影响机制,证实肠道菌群差异具有表观特征,可通过中医体质辨识证候分析得到临床表观证据预测肠道病毒重型易感人群,从而提高临床疾病预判及发挥中成药疗效。
