As a potential pathogenic factor of periodontal disease, virus has been given more and more attention. Its position in the occurrence and development of periodontitis is still not clear. In our previous research, we found that Anelloviridae is a major component of virome in periodontal environment and showed a higher abundance in chronic periodontitis group. The case control study revealed that Anelloviridae may be associated with chronic periodontitis. In this program, we will take TTMV-222 related periodontitis subjects and periodontal health subjects for research with the application of metagenomics, and analyse the difference in transcriptome, miRNome and circRNome. Bioinformatics method and luciferase report will be used to screen the circRNAs, interacted miRNAs and target genes related with chronic periodontitis. The pathogenesis of virus related periodontitis and molecular mechanism of inflammatory factors regulated by circRNAs after TTMV-222 infection will be explored. This program will provide a new perspective and theoretical basis for the etiological study of virus related periodontitis.
病毒作为一种潜在的牙周病致病因素逐渐受到关注,申请者前期研究发现细环病毒是牙周环境病毒群落的重要组成成分,慢性牙周炎患者细环病毒阳性率及病毒丰度显著高于对照组受试者,细环病毒可能与慢性牙周炎相关。细环病毒如何参与慢性牙周炎的发生发展尚待探索。本项目拟基于高通量分析,以细环病毒TTMV-222相关牙周炎患者和牙周健康者为研究对象,深度分析炎症与正常状态牙周膜上皮细胞转录组、miRNA组及circRNA组,探索circRNAs在细环病毒感染牙周炎症环境和牙周健康环境中的差异,通过信息学筛选及荧光素酶等筛选验证与炎症关联的circRNAs及其相互作用的miRNAs和miRNAs的靶基因,解析TTMV-222介导的潜在的circRNAs在病毒相关牙周炎发病机制中的作用机理,以期为病毒相关牙周炎的研究提供一个全新的视角,为病毒相关牙周炎机制研究提供理论依据和研究方向。
