hsMAD2 protein is a key component in mitosis spindle assembly chechkpoint in cell cycle control. We studied the expression and its role of hsMAD2 in leukemia and hepatocellular carcinoma, using Western blot, RT-PCR and gene transfection methods. Results were: 1.hsMAD2 expression was decreased in cases of leukemia, indicating that it correlated with leukemia pathogenesis; 2. hsMAD2 expression rates in hepatocelluar carcinoma cases were lower than paracarcinoma liver tissues and liver cirrhosis tissues; 3. Cochicine had a differential role on different leukemia cell lines and this might be related to p53 function; 4. hsMAD2 could suppress cell cycle in HepG3 cells. The results show that hsMAD2 gene plays an important role in leukemia and hepatocellular carcinoma, and may contribute to the clarification of the pathogenesis of leukemia and hepatocelluar carcinoma.
关卡控制保证细胞分裂过程中遗传信息复制和分离的准确无误。hsMAD2基因是有丝分裂关卡的重要组分,其改变会造成细胞分裂时染色体分离异常。我们发现hsMAD2是白血病相关基因。为明确其在白血病发病中的作用,我们拟通过对病例的检测、基因转染实验阐明它在白血病发病中的作用,即在染色体畸变和基因组不稳定性中的重要作用,并探讨其生化机制。
