Diabetes vascular lesions is one of the main causes of disability and death in diabetes patients. Angiogenesis barrier is the main pathological changes of diabetes vascular lesions. Based on the foundation of Supplementing Qi and activating blood circulation (Danzhijiangtang Capsule) has obvious therapeutic effect on the diabetic complications, this project respectively in the whole animal and cell levels studies the mechanism of Danzhijiangtang Capsule promoting angiogenesis. First of all, to establish acute left lower limb ischemia of diabetic rats model with intragastric administration of Danzhijiangtang Capsule, ELISA method to detect the VEGF, Ang1, Ang2 levels; Western blot method to detect the expression of rat gastrocnemius muscle VEGF, Akt, eNOS, Ang1, Ang2, Tie2 protein; to establish Danzhijiangtang Capsule fingerprint; to culture peripheral blood EPC of rats in vitro, Western Blot method and RT-PCR method to analysis the influence of drug containing serum on EPC VEGF/ Akt/ eNOS signaling pathway, Ang/Tie2 system and endothelial cell autophagy. To study the mechanism of prevention and treatment of diabetic complications by Supplementing Qi and activating blood circulation method. In order to provide theoretical suppor and experimental basis for supplementing Qi and activating blood circulation method regulating angiogenesis in the state of endothelial damage in diabetic rats in chinese medicine.
糖尿病血管病变是导致糖尿病病患者致残及致死的主要原因之一。血管新生障碍是糖尿病血管病变的主要病理改变。本项目基于益气活血法(丹蛭降糖胶囊)对糖尿病并发症具有显著治疗效果的基础上,分别以整体动物及细胞水平研究丹蛭降糖胶囊对血管新生作用及其机制。首先建立糖尿病大鼠急性左下肢缺血模型,给予丹蛭降糖胶囊灌胃, ELISA法检测血清中VEGF、Ang1、Ang2水平含量;Western blot法检测大鼠腓肠肌VEGF、Akt、eNOS、Ang1、Ang2、Tie2蛋白表达;建立丹蛭降糖胶囊指纹图谱;体外分离培养大鼠外周血EPC,采用Western Blot和RT-PCR的方法分析含药血清对EPC的VEGF/ Akt/ eNOS信号通路、Ang/Tie2系统及内皮细胞自噬的影响。为阐明益气活血法调控糖尿病大鼠内皮损伤状态下血管新生提供理论支持和实验依据。
糖尿病血管病变是导致糖尿病病患者致残及致死的主要原因之一。血管新生障碍是糖尿病血管病变的主要病理改变。本项目基于益气活血法(丹蛭降糖胶囊)对糖尿病并发症具有显著治疗效果的基础上,分别以整体动物及细胞水平研究丹蛭降糖胶囊对血管新生作用及其机制。在体水平上,实验采用自发性糖尿病大鼠(GK大鼠)建立了糖尿病大鼠急性左下肢缺血模型,给予丹蛭降糖胶囊干预。建立糖尿病大鼠右下肢缺血模型,结果显示,丹蛭降糖胶囊能够改善模型大鼠的精神状态及一般生理状况;能够改善下肢缺血 GK 大鼠的血糖、胰岛素、糖化血红蛋白和血脂水平,并且与用药剂量呈正相关;改善 GK 大鼠血液流变性指标;增加大鼠下肢新生血管密度,且呈剂量依赖性;降低GK 大鼠血清MMP-9 水平,促进血管基底膜的解离。体外实验中,分离GK大鼠股动脉内皮细胞体外培养,进行丹蛭降糖胶囊血清体外干预,结果显示:不同浓度含药血清体外干预 GK 大鼠下肢缺血细胞模型,丹蛭降糖胶囊含药血清减少内皮细胞凋亡,降低mTOR通路关键分子mTOR、s6k1、IGF1、4EBP1 mRNA表达,和mTOR、p-mTOR、s6k1、p-s6k1、p-IGF1、IGF1、p-4EBP1、4EBP1、IRS1、IRS2蛋白表达,升高细胞自噬相关分子beclin-1mRNA、ATG5 mRNA、LC3 mRNA和蛋白表达;增加了细胞出芽的面积与长度,结果表明丹蛭降糖胶囊调整糖尿病大鼠的血糖血脂代谢和胰岛素水平,还可以通过改善血液流变性,调控血管内皮损伤与新生相关的细胞因子的表达,能够抑制 mTOR 及其磷酸化过程,激活自噬的发生,促进自噬体的形成及血管出芽,具有促血管新生作用,其机制可能与mTOR/自噬信号转导通路有关。本项目为阐明益气活血法调控糖尿病大鼠内皮损伤状态下血管新生提供理论支持和实验依据。
