White Spot Syndrome Virus (WSSV) is currently the most serious shrimp pathogen, which has brought huge losses to shrimp industry worldwide. Until now, there is no effective ways to control WSSV because of complexity of pathogenic mechanism. WSSV had emerged the variation between different strains accordingly. The differences between different isolates may lead to WSSV virulence variation. Clarifying the molecular mechanism of genetic variation and pathogenesis of the WSSV will benefit to elaborate the pathogenic mechanism and to adopt the epidemic prevention strategy..The project will focus on the key scientific problem of variation and pathogenic differences. Based on the sequencing of different virulence WSSV and expressing of deleted gene, the role of diversity gene in WSSV infection and molecular response of shrimp to different virulence WSSV will be explored by methods of pathogen biology, bioinformatics and molecular biology. The difference of variation and pathogenesis will be investigated. Furthermore, the effect of diversity gene on pathogenesis will be clarified. Additionally, virulence associated gene and the intercome-molecules will be selected. The research will provide theory evidence for pathogenic molecular mechanism, antiviral drug target and prevention and control strategy for WSSV.
对虾白斑综合征病毒(WSSV)是对虾的主要病原并造成对虾业的严重损失。虽然国内外学者对WSSV进行了大量的研究, 但由于该病致病机理的复杂性, 至今仍无有效的防制方法。由于WSSV具有易变异的特性, 因而对其基因组进行遗传变异及致病力差异分析, 对于阐明其致病机理及制定相应的防疫战略具有重要的意义。.本课题围绕WSSV变异与致病力差异关键科学问题,在前期不同毒力毒株基因测序、差异基因分析与表达的基础上,将病原生物学、生物信息学与分子生物学相结合,从病毒差异基因在病毒感染中的作用及宿主对不同毒株分子响应等方面, 深入研究WSSV变异与致病力差异的分子机制,明确差异基因对WSSV致病力的影响,筛选WSSV毒力相关基因及其与宿主互作的分子,为WSSV致病的分子机理、药物靶标筛选及WSSV防控策略的完善提供理论依据。
对虾白斑综合征病毒(WSSV)是对虾的主要病原。由于WSSV具有易变异的特性,因而对其基因组进行遗传变异及致病力差异分析, 对于阐明其致病机理及制定相应的防疫战略具有重要的意义。本课题围绕WSSV变异与致病力差异关键科学问题, 从病毒差异基因在病毒感染中的作用及宿主对不同毒株分子响应等方面,深入研究WSSV变异与致病力差异的分子机制。解析wsv001、wsv006、wsv089、wsv150、wsv313在不同地区样本蛋白结构变异的分子特征, 包括氨基酸大小片段缺失、氨基酸插入、氨基酸突变。运用核酸组学和差异蛋白组学方法, 研究了WSSV强弱毒株对宿主miRNA和蛋白表达谱以及信号通路相关基因表达的影响, 初步阐明不同毒力毒株对宿主致病性和信号通路激活能力差异。采用酵母双杂交的方法筛选出与变异基因互作的宿主靶分子多个,分析互作靶分子对WSSV感染及相关信号通路的调控作用。发表论文12篇 (其中SCI期刊论文4篇),培养硕士研究生4名。本课题研究结果为WSSV致病的分子机理、药物靶标筛选及WSSV防控策略的完善提供理论依据。
