Dodder is a most common annual parasitic weeds of soybean,which is a worldwide weed quarantine object. Regrowth characteristics of isolated dodder have not been explored which led to exacerbate prevention difficulty in soybean flied. In the present project, morphology and molecular biology characteristics would be studied in isolated mature stem segments of dodder (Cuscuta campestris), and programmed cell death (PCD) sites and features would be explored. The starch grains in spatial and temporal distribution characteristics would be detected in the bottom and top of dodder stem segments, activities of SuSy,HXK, and Inv would be tested and soluble sugars (sucrose and fructose) metabolism and transfer characteristics would be explored. Assimilation capacity of synthesis of isolated dodder would be proved by 14C feeding method, and energy consumation (respiration) were explored. Dodder cDNA library would be constructed, and analyzed transcriptome sequence information. The expression levels of starch synthase, invertase enzyme and sucrose synthase gene should be detected by RT-PCR or qPCR in dodder stem segments. Silenced starch synthesis enzymes and sucrose-converting enzyme by inhibitors would be used to prove assimilates transport role in the regeneration process in the Dodder. The results would reveal dodder regeneration provide the theoretical basis, and supply the theoretical basis for the material transportation characteristics of higher plants under nutrient deficiency.
菟丝子是大豆田间常见的一年生恶性寄生杂草,是世界性的杂草检疫对象,其具有脱离寄主再生长特性,加剧了其防止难度。但有关离体菟丝子再生长机制缺乏深入研究。本项目以田野菟丝子脱离寄主成熟茎段为材料,在黑暗和光照条件下,研究其再生过程中形态学下端程序细胞化死亡(PCD)发生的形态学与分子生物学特征;分析离体菟丝子不同区段内淀粉、蔗糖等合成与降解动态、糖代谢关键酶SuSy、Inv等活性、光合作用及能量消耗等动态变化,探讨离体菟丝子碳同化产物代谢特征;应用菟丝子转录组数据库和糖关键酶抑制剂等,分析离体菟丝子SuSy、HXK和Inv等基因时空表达特征分析,揭示离体菟丝子形态学下端PCD和上端茎尖生长过程糖信号表达的转导途径。研究结果对揭示菟丝子再生长特性提供了理论基础,为有效防治离体菟丝子再侵染提供了理论依据,对研究高等植物在养分亏缺下同化物转运规律具有重要的理论意义。
