Electro-acupuncture at "Quchi (LI 11)", "Zusanli(ST 36)" improves the motor function after cerebral apoplexy, the clinical curative effect is exact, and it has been widely used. The preliminary affirmed the brain protective effect of electro-acupuncture in improving the neurologic deficit, from the aspect as follow: RA signaling pathways regulated endogenous NSCs proliferation, the calcium regulation by G protein, potassium channel inhibitted the release of the neurotransmitter GABA, etc. But all of these confined to in vitro, local, static, isolated brain regions, and the mechanism of the central nervous system was not very clear. This study intends to use rats with MCAO model as the research object, by using the 7.0 T fMRI to observe the MRI signal changes of cerebral areas when electro-acupuncture at "Quchi (LI 11)", "Zusanli(ST 36)",simultaneously combined with transmission electron microscope to observe the morphological changes of synapses, from the view point of whole, function, dynamic to discuss the central nervous system mechanism of electro-acupuncture at "Quchi ", "Zusanli" improving dyskinesia after stroke, at the same time provide the visibility experimental basis, and provide theoretical basis for clinical clinical application.
电针“曲池”、“足三里”改善脑卒中后运动功能,临床疗效确切,已被广泛应用。前期研究从RA信号通路调控内源性NSCs增殖及G-蛋白调控钙、钾通道抑制神经递质GABA的释放等方面,肯定了电针改善神经缺损功能的脑保护作用,但局限于离体、局部、静态、孤立的脑区,且其中枢作用机制尚不十分明确。本研究拟以MCAO模型大鼠为研究对象,采用7.0T功能核磁( fMRI)观察电针“曲池”、“足三里”脑区的核磁信号变化,从整体、功能、动态的角度探讨电针“曲池”、“足三里”改善脑卒中后运动功能障碍的中枢机制,同时结合透射电镜观察电针“曲池”、“足三里”对神经突触形态学的影响,进一步提供可视性的实验依据,为临床应用治疗提供理论基础。
临床证实电针“曲池”、“足三里”改善脑卒中后运动功能障碍疗效确切,有文献资料通过动物实验肯定了电针改善神经缺损功能的脑保护作用,但其中枢作用机制尚不十分明确。本课题制备MCAO模型鼠模拟临床急性脑梗死病理,对MCAO模型大鼠进行电针“曲池”、“足三里”干预,观察大鼠行为学(神经行为学Zea-Longa评分、Catwalk指标、Homecage指标)、功能核磁信号、脑组织超微结构及突触表达之间的关系,结果发现电针“曲池穴”、“足三里穴”可明显改善大鼠的神经行为学评分,提高大鼠的步行能力以及日常行为如进食、舔毛等,缩小了大鼠的脑梗死体积;可对不同脑区产生激活点,并改善大鼠脑组织的超微结构,增加突触的数量,上调可突触相关蛋白Synaspsin、PSD95的表达,且具有一定的时间依赖性。
