Microwave has been widely used in the process of producing and living, and the effect of microwave on human health has been recognized gradually. Hepatic injury includes the injury to liver from diverse factors. But the precise causes and mechanisms of it were still unclarified. The studies about hepatic injury from microwave radiation were still very few. In present study, we aim to construct the rats model which has been irradiated by high powered microwave. Then, we will evaluate the association between the liver function of models and the microwave radiation. Further more, we will screen microRNAs which were expressed differentially in the primary liver cells in rat models by microRNA array. Moreover, we will collect the blood sample of the technicists before or after they participated in the aerospace controlling work. The microRNA expression will be detected in these samples by real-time PCR and the key microRNA will be determined. At last, we will stuty the effect of miRNA which were related with liver injury to the function of primary live cells by lentivirus overexpressing strategy and knockdown strategy in vitro. All of these results would find miRNAs which were related with liver injury from microwave radiation, and would provide novel idea and direction to defend and reverse the liver injury from microwave radiation.
微波广泛应用于生产和生活,其对人体健康的影响也逐渐得到认识。肝损伤是各种原因所致的肝脏损伤的总称,但其病因和发病机制尚未完全阐明,由高功率微波辐照所致的肝损伤尚未得到证实。本项目拟建立高功率雷达微波辐照的大鼠模型,通过全自动生化分析仪检测大鼠模型的肝脏功能,以评价微波辐照对大鼠肝脏功能的影响。随后,原代培养大鼠模型的肝细胞,提取RNA进行miRNA芯片的筛选,筛选出差异表达的miRNA。收集200例技术人员参加测控任务前后的静脉血,通过real-time PCR方法检测筛选出的miRNA的表达,以验证并确定微波辐射所致肝损伤相关的关键miRNA。采取慢病毒表达载体过表达或miRNA海绵抑制其表达的方法,体外研究肝损伤相关miRNA对原代肝细胞功能的影响,筛选出肝损伤相关的miRNA,为预防和逆转微波辐照所致的肝损伤提供新的思路和方向。
