The abuse of new designer drugs such as Methamphetamine, MDMA, and Ketamine has caused severe damage to the society, like drug driving. Saliva screening is an important approach for spot testing of drug abuse. Saliva matrix is pretty complex and the drug content is usually at a level of ng/ml. The widely used screening method immunoassays usually suffer from insensitivity and false positive response. This application is going to employ step voltage paper spray-high resolution ion mobility spectrometry for developing a fast spot testing method for illicit drugs of abuse in saliva. Step changing voltage and chromatography paper would be used to build a step voltage paper spray source for ionizing and preliminarily separating the saliva sample at the same time. An efficient interface for connecting the paper spray source and the high resolution ion mobility spectrometer would be built up to completely desolvate the ions from the paper spray source and efficiently transmit them to the high resolution ion mobility spectrometer. Ions travelling in the high resolution ion mobility spectrometer would be further separated, thus the target ions could be selectively and sensitively detected. The total time consuming of this method is less than 20 seconds, and the limit of detection could reach 20 pg/µl. This project would provide a reliable spot testing technology for the abuse of illicit drugs, especially drug driving.
新型毒品冰毒、摇头丸、K粉等的滥用引发社会矛盾和造成危害,如毒驾,唾液分析是目前现场检测毒品的重要依据。唾液基质复杂,毒品含量低(ng/ml量级),目前免疫检测方法在实际应用中灵敏度不够,而且常常造成假阳性响应。本申请拟采用阶梯电压纸喷雾电离-高分辨离子迁移谱技术,发展一种现场快速检测唾液中毒品的方法。本研究将利用色谱纸作为纸喷雾离子源的基材,并施加阶梯变化纸喷雾电压,对唾液进行离子化的同时对目标毒品做初级分离;发展纸喷雾离子源与离子迁移谱的接口,实现对样品离子的完全去溶剂化和高效传输;利用高分辨离子迁移谱对目标毒品离子进行第二级分离和检测,从而实现对样品中目标毒品的现场快速准确测定。整个检测时间小于30 s,检测限达到20 pg/µl。本项目将发展的纸喷雾-迁移谱测量方法,将为新型毒品的现场稽查,特别是毒驾提供新手段。
新型毒品冰毒、摇头丸、K粉等的滥用引发社会矛盾和造成危害,如毒驾。唾液分析是目前现场检测毒品的重要依据。唾液基质复杂,毒品含量低(pg/uL量级)目前免疫检测方法在实际应用中灵敏度不够,而且常常造成假阳性响应。针对这一问题,本研究基于纸喷雾离子源和离子迁移谱技术便携、适于现场检测的特点,通过研制纸喷雾离子源与高温离子迁移管的接口,搭建了用于现场检测唾液中毒品的纸喷雾离子迁移谱仪器平台。通过研制场切换结合离子门的高效离子萃取技术,解决了纸喷雾离子源产生离子的高效利用问题,同时不损失离子迁移谱的分辨能力;通过研制无离子歧视效应的TPG离子门,将离子迁移谱的分辨能力首次提高到120,解决了唾液中基质对目标毒品离子检测的干扰问题,同时保证了仪器对迁移率普遍较小的毒品离子的灵敏度。整个系统检测毒品的时间在1-1.5min之间,对冰毒的检测限达到800 pg/µl。本研究发展的纸喷雾超高分辨离子迁移谱测量方法,将为新型毒品的现场稽查特别是毒驾提供一种手段。
