Hyperuricemia is a serious metabolic diseases, however the incidence of which is become higher and higher in the past decades. Chicory is well known in Uighur folk medicine which can lower uric acid by combine many binds side for its complex chemical composition. Our team have find out that the components extracted from the root of chicory can decrease uric acid level, but the material foundation and its action pathway are not clear yet...In this research we want to investigate the possible material foundation and the function way of chicory with the spectrum-activity relationship theory by analysis the relationship of HPLC fingerprint of the root of chicory and its lower uric acid function. First of all, our team try to clarify how the chicory to lower uric acid and its character by study how the uric acid produce and how its excretion. Secondly, we want to establish the fingerprint of the compounds which are absorbed is blood by HPLC-MS method. Thirdly, we try to work out the possible material foundation and the function way of chicory by establishing and analysis the spectrum-activity relationship model of chicory. This research have significance mean both in academic value and clinical value for its set the foundation to work out how chicory lower uric acid and it’s also provide a new way to find out how TCM to lower uric acid.
高尿酸血症是一种全身慢性代谢性疾病,近年来患发病率升高危害严重。中药菊苣根有较好的降尿酸作用,但是其化学成分复杂,可能具有多个作用靶点。课题组在前期研究已经发现菊苣根能够干预尿酸的合成与排泄环节,但其物质基础与药效机制尚未完全阐明。. 本项目基于谱效相关性原理深入分析菊苣根化学指纹图谱与降尿酸的作用环节,阐明菊苣治疗高尿酸血症的物质基础与药效机制。实验采用HPLC-MS技术建立菊苣提取物及其入血成分的指纹图谱;同时动态观察菊苣提取物的药效特点,从尿酸合成与尿酸排泄途径分析菊苣对整体动物模型及体外细胞模型尿酸代谢通路的影响;进而采用生物信息学手段,建立谱效关系的数学模型,解析菊苣指纹图谱与药理效应的关联关系。课题将阐明菊苣治疗高尿酸血症的物质基础与药效机制,为菊苣治疗高尿酸血症的应用奠定实验基础,为高尿酸血症的中药防治提供新思路。课题具有较好的学术价值与临床现实意义。
