Abstract: By constructing the plasmids of DNA-BD/LMP1-PGBT 9 and DNA-AD/cDNA GADGH, We screened some clones with positive Gal4. Connecting with the sequential analysis, We have investigated these function and attribute. By analysizing the difference of cDNA in nasopharyngeal carcinoma cells with positive EB virus-LMP1 mutant and negtive mutant, We explored the influence of the EB virus-LMP1 to the gene expressing of nasopharyngeal carcinoma cells. We have got more than 10 gene fragments, of which some were found to have connection with regulating the proliferation and transformation of cells. By activating NFkB, the EB Virus-LMP1 regulates the relative gene expression, which plays an important role in the process of nasopharyrgeal carcinoma. The investigation will provide important experiment basis and theoretic support to the biological treatment to nasopharyngeal carcinoma.
EB病毒编码跨膜蛋白LMP1,其能调控感染细胞增生转化和恶变,并在鼻咽癌的发生过程中起重要作用。我们从中国人的鼻咽癌细胞中分离到特异的LMP1突变体,其比LMP1原型具有更强的致瘤性和更低的免疫原性。我们利用此突变体探讨其调控鼻咽癌细胞基因转录过程的时空效应,并寻找其下游结合蛋白,以进一步研究其癌机制和应用价值。
