Pigment epithelium derived factor (PEDF) is a multifunctional protein exhibiting a wide range of antioxidant, antitumorigenic, neurotrophic and vasopermeability inhibiting properties. According to our previous research, PEDF could promote the blood labyrinth barrier (BLB) component cells' growth and restore the damaged BLB component cells caused by acoustic trauma. Our preliminary experiment showed that there was obvious degeneration in the BLB component cells of presbycusis mice. So far, little is known about the effects of PEDF on the degeneration of BLB in presbycusis. So we address the hypothesis that PEDF can delay the hearing loss through delaying the BLB component cells degeneration and maintaining the BLB integrity. To verify this hypothesis,we will adopt such methods as immunohistochemistry, Elisa, real time PCR, western blot, TEM, and audiological examination on the C57BL/6 presbycusis mouse model to figure out the effects of PEDF in delaying the hearing loss of presbycusis from multiple aspects of molecule, cellula, tissue as well as the whole animal, specifying a mechanism that PEDF works on the maintenance of BLB. From a new sight of PEDF, this study will lay the foundation of revealing its mechanism of maintaining the BLB integrity. And it may also open new avenues for delaying and protecting against presbycusis.
色素上皮衍生因子(PEDF)是一种多功能蛋白,具有抗氧化、抗肿瘤、神经营养及抑制血管渗透等特性。我们前期实验结果显示PEDF能促进小鼠血迷路屏障(BLB)组成细胞生长,对噪声性聋小鼠的BLB组成细胞损伤有修复作用;预实验结果提示老年性聋小鼠BLB细胞有明显的退行性改变。迄今PEDF对老年性聋BLB退变的影响研究甚少。故我们提出假说:PEDF可能通过延缓BLB组成细胞退变,维持血迷路屏障完整来延缓听力损失的发生。为验证该假说,我们通过C57BL/6老年性聋小鼠模型,采用免疫组化、Elisa、real time PCR、western blot、透射电镜及听力学检测等手段,从分子、细胞、组织及动物整体水平多方面探讨PEDF维持血迷路屏障完整性的机制及其在延缓老年性聋听力损失中的作用。本研究将从PEDF这个新视点为揭示其维持老年性聋血迷路屏障完整性的机制奠定基础,为延缓及预防老年性聋提供新思路。
