Epilepsy is a common and diverse set of chronic neurological disorders characterized by seizures.There is none efficient treatment way to cure the disease so far, currently, some study shows that Et-DHA has the effect of anti-epilepsy, but the mechanism does not be cleared. This project aims to investigate the neuroprotective effects of Et-DHA in the Kainic Acid-induced rat's adrenal pheochromocytoma cell line (PC 12 cells) and the possible mechanism.In our preliminary experiments,the results showed that pretreatment with Et-DHA significantly reduced LDH release, ROS production(P < 0.01)and cut down the activation degree of Caspase-3. Et-DHA protected PC12 cells against KA-induced apoptosis and oxidative stress injury via inhibition of mitochondrial dependent apoptosis pathway. Therefore, the present study is going to explore Et-DHA, a promising neuroprotective agent, for the new treatment strategy and new target of epilepsy in clinical medicine plus the mechanism of it's effect.
癫痫(Epilepsy)作为神经系统常见疾病,是慢性反复发作短暂脑功能失调综合征,目前还没有一种较有效的治疗策略。研究表明二十二碳六烯酸乙酯((Et-DHA)对癫痫具有一定保护效应,但其具体的作用机制尚未完全明确。本项目拟在PC12细胞以及癫痫动物模型上探讨Et-DHA对由红藻氨酸(KA)诱导的氧化应激损伤模拟的癫痫模型的保护作用及其可能的信号通路途径及分子机制。预实验结果表明:Et-DHA对PC12细胞具有明显的保护效应,显著提升细胞存活率(P < 0.01),减少乳酸脱氢酶(LDH)和活性氧簇(ROS)释放量,降低 Caspase-3活性,提示Et-DHA的保护效应可能通过抑制线粒体依赖凋亡途径抗氧化应激实现。本项目意在明确DHA对癫痫脑神经元保护的细胞信号转导途径及离子机制,从而进一步探索防治癫痫等神经性疾病的有效制剂和机理,以寻求预防和治疗癫痫新策略、新靶点,以更好地服务于临床。
本项目基本完成预期的研究计划,从细胞水平PC12、KA诱导的SD大鼠癫痫脑模型,利用western blot, RT-PCR, Pulse-chase metabolic labeling of Caspase proteins)等方面详尽的探讨了Et-DHA后处理,主要通过保护线粒体功能、降低ROS生成、抑制caspase凋亡通路(膜电位MMP、通透性)功能调控,调控A型钾通道Kv4.2电流及其调控蛋白KChIP1的表达,以及Et-DHA后处理通过调控丙二醛(MDA)、GSH(还原型谷胱甘肽)和GSSG(氧化型谷胱甘肽)水平、GPx and GRx 酶活性等,从而对SD大鼠癫痫模型的改善作用及对海马神经元的病理改善作用等组织学分析诸方面,改善癫痫症状效应,实现对癫痫脑神经元的保护效应。基本达到预期的研究结果,对临床辅助用药以及后期处理具有较好的指导意义。部分研究结果已在国内核心和SCI发表,有的结果正在审稿中,同时也申请了2项相关专利。另外我们还进一步对Et-DHA对神经干细胞调控改善SD大鼠癫痫模型做了探索。
