Metastasis is a key issue in cancer research. The ultimate goal of tumor metastasis mechanism research is to find drugs that can effectively inhibit metastasis. Triple negative breast cancer is a highly invasive subtype of breast cancer, and it is urgent to find effective clinical drugs. Arsenic trioxide(ATO) is a traditional medicine that has a significant effect on acute promyelocytic leukemia, and has been widely used in clinical treatment of leukemia. The applicant previously found that ATO and the BRD4 inhibitor JQ1 have a good combination effect in pancreatic cancer. The applicant's recent exploration found that ATO can significantly inhibit the metastasis of triple negative breast cancer cells to the lung. Preliminary analysis found that, on the one hand, ATO mainly inhibited the downstream gene expression of the TGF-β signaling pathway by inhibiting the phosphorylation of SMAD2 / 3; on the other hand, the transcription factor CEBP-β Plays an important role in inhibition of ATO on TGF-β signaling. This project intends to further study the function and mechanism of ATO to inhibit breast cancer lung metastasis by regulating CEBP-β and TGF-β signaling pathway, and to explore its combination effect with TGF-β pathway inhibitor in breast cancer lung metastasis, to provide a new strategy for the clinical treatment of lung metastasis in breast cancer.
转移是癌症研究的关键问题。肿瘤转移机制研究的最终目标是寻找可以有效抑制转移的药物。三阴性乳腺癌作为乳腺癌中具有高侵袭性的一大亚类,目前急需寻找有效临床药物。三氧化二砷是一种对急性早幼粒白血病的疗效特别显著的传统药物,已在临床上白血病的治疗中被广泛使用。申请人前期发现三氧化二砷与BRD4抑制剂JQ1在胰腺癌中具有良好的联合用药效果。申请人最近的探索发现砷剂可以明显抑制三阴性乳腺癌细胞向肺部的转移过程。前期初步分析发现,一方面砷剂主要是通过抑制TGF-β信号通路下游转录因子SMAD2/3的磷酸化来抑制该通路下游基因表达;另一方面转录因子CEBP-β在砷剂对TGF-β的抑制作用中起到重要的调控作用。本项目拟进一步研究砷剂通过调控CEBP-β与TGF-β信号通路抑制乳腺癌肺转移的功能及其机理机制,并将探索其与TGF-β通路抑制剂在乳腺癌肺转移中的联合用药效果,为乳腺癌肺转移的临床治疗提供新的策略。
