Hyperplasia of mammary glands is the clinical common and frequently-occurring disease. Estrogen and progesterone is closely related to development of the disease. Based on clinical and experimental early stage of the Xihuang Pill anti hyperplasia of mammary glands research, it is going to research this project with pathology, cell biology, molecular biology and immunology. Researching the influence patent traditional Chinese medicine Xihuang Pill on hypothalamus-pituitary-ovarian axis hormones and receptors expression level, the hypothalamus, pituitary, ovary ultrastructural organization structure and hypothalamic GnRH nerve cells, adenohypophysis cells, ovarian granulosa cells secretion in vitro, it plans to explore effect of Xihuang Pill on hypothalamus-pituitar-ovarian axis adjustment. By studying Xihuang Pill on mammary gland hyperplasia of mammary gland hyperplasia and related organs, animal model, related hormones and receptor expression level, the model of animal mammary gland hyperplasia tissue microenvironment and mammary gland epithelial cell growth and secretion influence, this project aims to clarify Xihuang Pill resistance mechanism of mammary gland hyperplasia. This project provides the experimental basis for Xihuang Pill of hypothalamus-pituitary-ovarian axis adjustment research and provides theoretical basis for the Xihuang Pill of mammary gland hyperplasia clinical application and breast cancer prevention.
乳腺增生是临床常见病多发病,内分泌紊乱、卵巢功能失调、雌激素、孕激素水平及活性与该病发生发展密切相关,囊性增生有癌变可能。西黄丸治疗乳腺增生病临床疗效确切。项目组前期研究发现,西黄丸具有抑制雌激素水平过高致乳腺增生作用。我们推测西黄丸可能通过调节下丘脑-垂体-卵巢轴抗乳腺增生,本项目应用病理学、细胞生物学、分子生物学、免疫学技术,通过研究传统中成药西黄丸对下丘脑-垂体-卵巢轴各激素及受体表达水平,下丘脑、垂体、卵巢超微组织结构和离体下丘脑GnRH神经细胞、腺垂体细胞、卵巢颗粒细胞分泌功能的影响,探讨西黄丸对下丘脑-垂体-卵巢轴的调节作用;通过研究西黄丸对乳腺增生组织及相关脏器、模型动物乳腺增生相关激素及受体表达水平、模型动物乳腺增生组织微环境和乳腺上皮细胞生长及分泌功能的影响,阐明西黄丸调节下丘脑-垂体-卵巢轴的抗乳腺增生作用机制,为西黄丸抗乳腺增生临床应用及乳腺癌防治提供理论依据。
