The chromosomal passenger complex (CPC) plays an important role in cell division. Other studies have shown that Ubiquitin specific peptidase 22 (USP22) as a type of deubiquitinase regulates the expression of multiple tumor-related genes. In our previous work, we found that PARP6 regulated the expression of CPC, and high expresson of CPC was associated with a poor prognosis in OSCC.The further results show that Usp22 is highly expressed in OSCC, and USP22 siRNA reduced CPC expression in OSCC cells. Based on the aboved information, we hypothesize that USP22 may regulate the expression of CPC protein by deubiquitination, and promote the occurrence and development of OSCC. This project will clarify the mechanisms of USP22 regulate CPC and promote the occurrence and development of oral cancer.The project will clarify the mechanism of how USP22 regulate CPC and promote the occurrence and development of oral cancer. It is providing clues for the occurrence and development of oral cancer and is important in the prognosis judgement, gene diagnosis and treatment in cancer. It is providing theoretical basis and experimental basis in the early diagnosis, prognosis determination and treatment of OSCC.
染色体乘客复合体蛋白(CPC)是一种与细胞分裂密切相关的蛋白。已有研究表明泛素特异肽酶22(USP22)作为去泛素化酶,可以调控多种肿瘤相关基因的表达。我们前期的工作发现:在口腔鳞状上皮细胞癌(OSCC)中CPC的高表达与OSCC恶性程度密切相关,并且CPC蛋白的表达水平受到USP22的调控。进一步结果显示:USP22在OSCC中高表达;下调USP22在OSCC细胞中的表达,CPC蛋白的表达也随之降低。据此我们推测,USP22可能通过去泛素化作用调控CPC蛋白的表达水平,进而影响OSCC的发生发展。本项目旨在明确USP22如何调控CPC蛋白,及其在口腔癌发生发展中的作用及分子机制。为OSCC的早期诊断、预后判定及治疗等方面提供理论依据和实验基础。
