AP311, a bacteriocin produced by Lactobacillus acidophilus ,has a broad spectrum of activity against Gram-positive bacteria and Gram-negative bacteria,especially some pathogenic bacteria. In this study, AP311 was purified to homogeneity by n-propanol precipitation, sequential SP-Sepharose Fast Flow, phenyl-superose and Sephadex G-25. According to its N-terminal amino acid sequence, we synthesized a pair of oligonucletide primers and amplified successfully the genes involved in AP311 biosynthesis from Lactobacillus acidophilus total DNA. Amino acid and nucleotide sequencing revealed AP311 is a novel bacteriocin, it compprises a 33-amino-acid peptide with an amino acid sequence similar to that of Salivaricin A.
从德氏乳杆菌分离到的抑菌活性物质AP-311是一种未见文献报道的新的广谱抗菌多肽,它不仅对革兰氏阳性菌、而且对革兰氏阴性菌,特别对某些致病菌有不同程度抑制作用。本项目以AP-311 N-端氨基酸序列合成寡核苷酸探针从德氏乳杆菌总DNA中分离该抗菌多肽的生物合成基因,并进行DNA全序列分析,并研究其在乳酸菌系统中的功能表达。
